Alendronate was presented by Merck Sharp &Dohme S.p.A. Collagenase-Ⅱand Trypsin were obtained from Invitrog. FBS was obtained from cell scinence.Methods:①Osteoblasts isolated from Human ilium cancellous bone were treated with various concentrations of Alendronate (10-9~10-4M).②To investigate the effect of the concentrations of Alendronate(10-8M) on the biological characteristics of osteoblasts and osteoclasts in co-culture system.③MMT method was used to assay proliferation of osteoblasts. The osteoblasts were plated at the concentrations of 10-9~10-5 M, seventy two hours later treatment factors were set. Total RAN was extracted from osteoblasts using TRIzol , and mRNA expression of OPG and RANKL were examined using RT-PCR.Results:①10-5,10-4M alendronate inhibited proliferation of osteoblast; The proliferation of osteoblasts increased under the stimulation of Alendronate range 10-9~10-6M (P < 0.05),Peak levels were gained at concentration of 10-8M. Alendronate concentration-dependently inhibited RANKL mRNA expression,10-5M Alendronate has a maximum effect after 72 h of treatment( P<0.01).Alendronate concentration-dependently increased OPG mRNA expression,10-6M Alendronate has a maximum effect after 72 h of treatment( P<0.01).But it is reduced on 10-5M.②The proliferation and the ALP activity of osteoblasts increased by Alendronate in co-culture system. The bone lacuna value and the TRAP activity of osteoclast decreased by Alendronate in co-culture system.Conclusion:①Alendronate can increase the proliferation of osteoblast. The effect has a proper concentration of Alendronate.
