Objective: To explore the inhibitory effect on survivin mRNA expression and the proliferative and apoptotic effect in different positions siRNA targeting survivin in human androgen- independent prostate cancer PC-3 cells at the different time points.Methods: Used androgen-independent prostate cancer PC-3 cells as model, we constructed a multiple short hairpin RNA (shRNA) expression vector containing two shRNAs against survivin and two single shRNA vector containing only one survivin shRNA, which were transfected into the PC-3 cells by liposome method. Then transfection efficiency was observed under fluorescence microscope. PC-3 cells were divided into 5 groups: survivin 1 group, survivin 2 group, survivin 1+2 group, negative control group and cell control group. At 24 h, 48 h and 72 h after the transfection, the expression of survivin mRNA in PC-3 cells was tested respectively by half-quantity RT-PCR.The proliferation of PC-3 cells was detected by MTT assay. Apoptotic rate was detected by Annexin V-PE-FCM assay.Results: Compared with the control groups, survivin 1 group, survivin 2 group and survivin 1+2 group inhibit the expression of survivin mRNA at 24h after transfection. The highest inhibition ratio occurred at 48 h after transfection, which was 52%.And after 72 h the ratio descended. All the groups targeting survivin efficiently suppressed the proliferation of PC-3 cells. survivin 1 group had the most powerful effect and all the groups can induce cells apoptosis, survivin 1 group had the highest apoptotic rate of 13.6%±1.8% at 48h after transfection.Conclusion: After the transfection, siRNA can inhibit he expression of survivin mRNA, can effectively inhibit cell growth and induce cell apoptosis in androgen- independent prostate cancer PC-3 cells. It suggest that RNAi targeting survivin has a potential value in the gene therapy. The experiment show that he highest apoptotic rate at 48h after transfection.And siRNA, which aimed at different positions of survivin gene, has different interfering effects.
