Objective To investigate Change of serum CRP and Mean alveolar number(MAN)Pulmonary mean linear intercept(MLI ) of Emphysema Guinea Pig model when they have been Managed With Long-Acting ?2-Agonists and Inhaled Corticosteroids Combination ,to provide basis for Management of Emphysema With Long-Actingβ2-Agonists and Inhaled Corticosteroids Combination.Methods twenty-one male Guinea Pigs and twenty-one female Guinea Pigs were randomly divided into five groups , A group group was control group(n=9), B group was no treatment group (n=9), C group was ICS treatment group(n=9),D group was LABA treatment group(n=9),E group was LABA/ICS treatment group(n=9).The emphysema Guinea Pig model was established by exposed to the smoke of 10 cigarettes for 1-1.5hours twice. After the emphysema Guinea Pig model was established, Guinea Pigs in B group were not treated for a month, Guinea Pigs in C group were treated with ICS for a month ,Guinea Pigs in D group were treated with LABA for a month ,Guinea Pigs in E group were treated with LABA and ICS combination for a month . After the study was complished, the Guinea Pigs were exsanguinated after we obtained blood samples from the hearts of these Guinea Pigs, lung block from the pulmonary lobes was embedded in paraffin, and was stained with hematoxylin-eosin,Calculating Pulmonary mean linear intercept (MLI) and Mean alveolar number (MAN), Serum CRP of Guinea Pigs was measured by enzyme-linked immunosorbent assay (ELISA).Results1.The Guinea Pigs were exposed to cigarette smoke whose Air spaces were enlarged and Some alveoli were confluent,the pathological findings reached the standard of emphysema model.2.The serum CRP levels of emphysema Guinea Pigs model were significantly higher than that in the control group(p<0.05). the serum CRP levels of H group were higher than that in the B group, the serum CRP levels of B group were higher than that in the C group, the serum CRP levels of C group were higher than that in the A group.but the serum CRP levels had no significant difference among four groups of emphysema Guinea Pigs model.3.MLI in the four Emphysema Guinea Pigs model groups had a significant increase than in comparison with the control group(p<0.001). MLI in the H group had increase in comparison with the B group, but MLI had no significant difference between them. MLI in the B group had a significant increase than in comparison with the C group(p<0.001). MLI in the Cgroup had a significant increase than in comparison with the A group(p<0.001).4. MAN in the four emphysema Guinea Pigs model groups had a significant decrease than in comparison with the control group(p<0.001). MAN in the H group had decrease in comparison with the B group, but MAN had no significant difference between them. MAN in the B group had a significant decrease than in comparison with the C group (p=0.018). MAN in the C group had a significant decrease than in comparison with the A group(p<0.001).5.We found there were significant negative correlation between MAN and MLI ,r=-0.824 (p<0.001).there were negative correlation between MAN and CRP concentration, r=-0.572(p=0.001). there were positive correlation between MLI and CRP concentration ,r=0.458(p=0.011).Conclusions1.The serum CRP levels of emphysema Guinea Pigs model were significantly higher than that in the control group(p<0.05). The Guinea Pigs treated with LABA/ICS combination therapy had no significant reductions in serum CRP levels compared with ICS or LABA alone therapy. 2. The Guinea Pigs treated with LABA/ICS combination therapy had a significant increase in MAN compared with ICS or LABA alone therapy. The Guinea Pigs treated with LABA/ICS combination therapy had a significant decrease in MLI compared with ICS or LABA alone therapy. The Guinea Pigs treated with LABA/ICS combination therapy had a trivialer emphysema compared with ICS or LABA alone therapy.
