Objective:To detect the expression of tumor suppressor gene P16 and HPV in tissue of normal uterine cervix,cervical intraepithelial neoplasia and cervical carcinoma, and to investigate relationship between expression of P16 and HPV with clinicopathologic parameters of cervical carcinoma. It is excepted that combined detection of P16 and HPV can be used for prevention,diagnosis and prognosis of cervical carcinoma.Methods:Using SP immuno- histochemical method, expression of P16 and HPV in patients of 30 normal uterine cervixes, 14 cervical intraepithelial neoplasias and 42 cervical carcinomas was retrospectively examined. Analysis were performed using the SPSS 12.0 statistical package. The association between expression of P16 and HPV with clinicopathologic parameters (such as age,tumor size,histologic grade,tumor infiltrate deepness,clinical stage,lymph node metastasis) was assessed by the X2 test or Fisher's exact test. Cox step wise regression model was used in multivariate analysis for prognosis evaluation. P<0.05 was significant statistically.Results:The expression of P16 protein and HPV in cervical carcinoma was higher than in normal cervical tissue (P<0.05). Exression of P16 protein was corelated with histologic grade and lymph node metastasis of cervical careinoma (P<0.05), but not relate to tumor infiltrate deepness,clinical stage,patients' age,tumor size (P >0.05). Exression of HPV in lymph node was corelated with clinical stage and lymph node metastasis of cervical careinoma(P<0.05), but not relate to tumor histologic grade and patients' age (P >0.05).There was corelationship between expression of P16 and HPV in cervical carcinoma.Conclusion:Expression of P16 and HPV was corelated with clinicopathologic parameters and prognosisof cervical carcinoma, and there was corelationship between expression of P16 and HPV incervical carcinoma. Combined detection of P16 and HPV can serve as an useful refer indexto judge lymph node metastasis,to predict prognosis and to select clinical treatment.
