BACKGROUND&OBJECTIVE: To investigate the relationship of high mobility group box protein1(HMGB1) and vascular endothelial growth factor C(VEGF-C) to the pathologic types, proliferation, lymph-node metastasis of lung carcinomas .METHODS: Immunohistochemical staining was performed to detect the over-expression of HMGB1 in 10 cases of small cell lung cancers samples, 42 cases of non-small cell lung tissue and adjacent tissue samples,ten samples of lymph-node with metastasis and 5 samples without metastasis. Six normal lung tissue were used as controls. VEGF-C was detected in all the non-small cell lung cancer with the same method.RESULTS: HMGB1 is over-expressed in 90%(9/10) of small cell lung cancers, 66.67% (28/42) of non-small cell lung cancers and 35.7%(15/42) of adjacent tissue samples. The difference of expression of HMGB1 between squamous-carcinoma and adeno- carcinoma is remarkable (45.0 % vs 86.36 %, P<0.05); also in their adjacent tissues (55% vs 18.18%, P<0.05). HMGB1 is much common over-expressed in NSCLC with positive lymph-node metastasis compared to those without lymph-node metastasis (94.44% vs 45.83%, P<0.05). There seems no relationship between the exprssion of HMGB1 and tumor differenciation. VEGF-C expression is detected in squamous-carcinoma and adeno-carcinoma (45% vs 81.81%, P<0.05) and there is a remarkable difference bwtween the samples with lymph node metastasis and those without lymph node metastasis ( 88.89% vs 45.83% , P<0.05). The exression of HMGB1 is positively correlative to VEGF-C.CONCLUSION: HMGB1 is over-expressed in lung cancers and related to pathologic types, lymph node metastasis. There seems a positive correlativity between HMGB1 and VEGF-C over-expression in NCSLC.
