[Objective] To investigate the expression and significance of ATP-binding cassette transporter (ABCG2) in renal carcinoma tissues and the relationship between the ABCG2 and renal carcinoma stem cell.[Methods] Expression of ABCG2 protein was examined by immunohistochemical technique in specimens from 65 renal carcinoma tissues( clear cell carcinoma) and 15 surronding normal tissues. The expression of ABCG2 mRNA was measured by RT-PCR in specimens from 30 renal carcinoma tissues and 10 surrounding normal tissues.[Results] The study confirmed the predominant localization of ABCG2 transporter in plasma membrane of the carcinoma cell , only little part was localization in cytoplasma of the tumor cell. ABCG2 protein expression was high in the renal carcinoma cell, while in normal renal tissues the expression was low or no expression. The positive rates of ABCG2 expression in Undifferentiated carcinoma was significantly higher than those in well and/or moderately differentiated carcinoma (P<0.05). The mRNA expression of ABCG2 was significantly higher in renal carcinoma tissue than in normal renal tissues(P<0.05).[Conclusion] The expression of ABCG2 in renal carcinoma tissue was high. ABCG2 may be closely related to the oncogenesis and progress of renal cancer, and may be an important factor in the research of renal cancer stem cell.
