Effect Of Flavonoids From Fruit Residues Of Vaccinium Vitis-idaea L. On Hyperuricemia And Pilot Product's Lowering Uric Acid Activity

In previous work,we established an extracting technique of flavonoids from fruit residues of Vaccinium vitis-idaea L.,and studied on its further utilization industrially. Meanwhile,flavonoids from fruit residues of Vaccinium vitis-idaea L.made in lab (FVL) showed a supression effect to Xanthine Oxidase,which is an important enzyme in uric acid metabolism.In present work,we studied the lowering uric acid effect of FVL by using model of adenine-reduced hyperuricemia in rats.In this test,FVL obviously decreased serum uric acid in 1 week,and decreased it to normal level in 2 weeks.At the third week, FVL(100 mg/kg bw,200 mg/kg bw) obviously desreased XOD activity and serum uric acid,while all the dosage obviously decreased kidney index,urea nitrogen and malonaldehyde.Results showed that FVL could lower serum uric acid and protect kidney in adenine-reduced hyperuricemia rats.Since medium experiment had been industrialized based on our previous study,we compared flavonoids from fruit residues of Vaccinium vitis-idaea L.made in plant (FVP) valid composition and reducing uric acid effects with FVL.According to anthocyanidin interference of traditional method NaNO2-Al(NO3)3-NaOH measuring flavonol content in FVL;we used a new method called Three Wavelength Spectrophotometry,showed that FVP and FVL contained flavonol (reference to Rutin)15.02%and 13.07%,nearly to the results tested by HPLC 17.96%and 15.96%.In addition,we compared the lowering uric acid effects between FVP and FVL by using model of adenine-reduced hyperuricemia in rats.Results showed FVP could obviously reduce serum uric acid,kidney index,urea nitrogen and creatinine level. Further to observe the microstructure of kidney cells by transmission electron microscopy(TEM) showed FVP and FVL increased the quantity of mitochondria and crista,repaired the shape of karyon and mitochondria,contrasting with kidney cells of rats treated adenine:karyon irregular,nuclear pore blur,mitochondria and crista decreased,lysosome appeared,floss increased.
