Objective:By detecting the peak index of the main peak,the quality of the main peak,the average quality of DNA in the samples of prostate needle biopsy with image cytometry(ICM),to gain the average DNA ploidy value and the main peak DNA ploidy value. Combining S-phase fraction(SPF),G0G1 phase fraction(G0G1%), G2M phase fraction(G2M%) prostate cancer,to analysis the relationship between them and Gleason grade,clinical stage,in order to explore the value of the DNA ploidy analysis in diagnosis and therapy indirectly.Methods:59 cases of prostate needle samples in Huadong Hospital of Fudan University,of which 57 cases are prostate cancer, the other two cases high grade prostatic intraepithelial neoplasia (HGPIN).To detect the peak index of the main peak,the quality of the main peak,the average quality of DNA S-phase fraction (SPF),G0G1 phase fraction(G0G1%),G2M phase fraction(G2M%) with image cytometry(ICM).Results:sThe variance between the average DNA ploidy value and the different Gleason score groups is significant(F=3.943,p<0.05). The average DNA ploidy value and Gleason score have a positive correlation,while the main peak DNA ploidy value and Gleason score is not significantly correlated(r=0.433,p<0.01).The variance of the average DNA ploidy value in high middle low-risk prostate cancer groups is significant(F=5.742,p<0.01).The variance of the ratio of aneuploid,diploid of the average DNA ploidy in the low,middle high-risk groups is not significant.The variance of the main peak DNAploidy value the different Gleason score groups is significant.(F=1.201,p=0.318>0.05).The variance of SPF, G0G1%in the different Gleason score groups is significant,(p<0.05).The variances of the ratio of aneuploid,diploid of the average DNA ploidy in the low,middle high-risk groups are not significant(F=7.551,p<0.01).The average DNA ploidy value and clinical stage have a positive correlation(r=0.403,p<0.05).The variances of the average DNA ploidy value and the main peak DNA ploidy value between the groups which violate the capsule of the tumor or not are all significant.(the average DNA ploidy value F=16.916,p<0.001,the main peak DNA ploidy value F=4.921,p<0.05),especially the average DNA ploidy value.The diploid and aneuploid ratio of the average DNA ploidy change significantly between the two groups(p=0.003<0.01),But in the main peak DNA ploidy there are no significant differences (p=0.164>0.05).Conclusion:1.The average DNA ploidy value and Gleason score have a positive correlation,while the main peak DNA ploidy value and Gleason score is not significantly correlated.The variance of the average DNA ploidy value in high middle low-risk prostate cancer groups is significant.The average DNA ploidy value and clinical stage have a positive correlation.2.The average DNA ploidy value is one of the important mark of the prostate cancer which violate the capsule of the tumor or not. 3.The variance between the SPF and the different Gleason score groups is significant,but the SPF and the clinical stage has no correlation...
