OBJECTIVE: To investigate the compatible regularities of Radix Aconiti(RA) by HPLC and LD50 and clarify the chemical substances which could produce toxicity in the codecoction of Bawei Dihuang Recipe (BDR)and Ginseng Decoction(GD).METHODS: alkaoids in the Radix Aconiti were decocted in water and then were extracted, the diester-alkaloids and their changing regularities in the codecoction of BDR+GD, RA+GD, RA+Ginseng, RA+ Rhizoma Zingiberis(RZ) , RA+Radix Glycyrrh-izae(RG) were studied by HPLC; 5 to 7 dosage group were chosen at death rate from 0% to 100%, 10 mice each group, observed 7 days, then calculate the LD50 by Bliss.RESULTS: The diester-alkaoids in the codecoction of BDR+GD are most abundant, the LD50 is lowest and the toxicity of them is obvious; The diester-alkaoids in the codecoction of RA+GD, RA+AM are abundant, the toxicity of them is obvious; comparing with RA, the relevant alkaloids in the codecoction of RA+Ginseng basically unchanged, The relevant aconitine in the codecoction of RA+RG significantly reduced, and the toxicity degraded greatly, The relevant aconitine in the codecoction of RA+RZ are also more abundant, but the hypaconitine reduce Significantly, and the toxicity reduced greatly; RG and RZ can reduce the toxicity of RA significantly; AM can increase the toxicity of RA.CONCLUSION: Bawei Dihuang Recipe and Ginseng Decoction can indeed capable of producing toxic, the reason is that the hydrolysis of diester-alkaloids, like aconitine, in RA has been intensively restrained by AM in GD, which leads to the over high diester-alkaloids and induces the toxicity in the decoction of BDR and GD;RG and RZ can repel the toxicity of RA;Ginseng does not make the toxicity of the codecoction increase. |
