Studies On The Chemical Constituents, Extraction Process And Pharmacology Activity Of Uncaria Rhynchophylla (Miq.) Jacks

Uncaria rhynchophylla(Miq.)Jacks belongs to the Uncaria genus of Rubiaceae family.It distributes mainly in Jiangxi,Guangdong,Guangxi, Hunan and Yunnan Provinces.The hooks and stems of this plant have been used as an important traditional Chinese medicine for the treatment of headache,hyper- tension for hundreds of years. We investigated the chemical constituents from the hooks and steams of Uncaria rhynchophylla(Miq.)Jacks and ten compounds have been isolated .The structures of five of them have been identified by the physico-chemical , spectral means and also by comparison with some authentic samples and their standard spectras.They areβ-Sitosterol(Ⅰ),Ursolic acid (Ⅱ),Isorhynchophylline(Ⅲ),Rhynchophylline (Ⅳ),Daucosterol (Ⅴ). Taking the content of rhynchophylline as observation index,we have studied the extraction processs by the L9(34) orthogonal design.Through a series of tests,we found out that the best extraction condition was extracting two times,ten times 70% alcohol in volume of the raw material and two hours for the first time;eight times 70% alcohol and one and half hour for the second time.It was observed that the effect of extract of Uncaria rhynchophylla (Miq.) Jack [total alkaloids of Uncaria rhynchophylla (Miq.) Jack, rhynchophylline and Isorhynchophylline] on the SHR(spontaneous hypertention rat) blood pressure and the vascular remolding.The results indicate that the extract of Uncaria rhynchophylla(Miq.)Jack has the function of reducing the blood pressure and improving the vascular remolding.
