ObjectiveTo study the autophagy after acute spinal cord injury in rats,the changes and duration after ASCI,this thesis makes every effort to explore the effect of autophacy in the process of ASCI,which will have the Spinal cord Injury therapy get access to the new scientific references and target.Methods20 male wistars(250±20g) were divided into a normal group and a group with injure at randomly for the test of autophagosome.Another 50 wistars were put into normal group and groups with injures 2h,6h,24h,48h for the investigation of the expression of LC3(microtubule associated protein 1 light chain 3,MAP1-LC3).Each wistar was designed an ASCI model based on the improved Allen's method.With transmission between the normal group and groups with injures are compared to investigate the formation of autophagosome.The way of immunoblotting or western blot is applied to investigate the expression of the concerning protein LC3 of spinalcord tissue in the corresponding areas between the normal group and the groups of injures(groups of 2h,6h,24h,48h).ResultsSpinal cord injury is shown under the transmission electron microscope (TEM)..In the group with SCI,autophagosome appears,the quantity of lysosome is increased,karyotype is irregular mostly and the swollen cytochondriome is predominant.Immunoblotting or western blot shows that after SCI,autophagosome concerning protein LC3 is increased in 2h and it is increased particularly in 6h,and in 48h,there is still some expression in it.ConclusionsAutophagy is activited after acute spinal cord injury in rats,and the duration of it lasts 48hours at least.Autophagy plays a role in the process of the spinal cord injury to some extent. |
