Among the total,the OA patients have 30 cases,they are the process group.Their age was from 48~75.Their synovium was remained when they accepted the arthroscope operation.The control group was non-OA cases,also 30 cases.There are 4 cases of synovium syndrome,20 cases of menisci injury,6 cases of ACL or PCL injury.Their synovium was also remained when they accepted the arthroscope operation.The research methodThe synovium were deal with NS water flush in one hour after the operation,to be cut by 1.0cm×0.8cm×0.5cm,then they were deal with the process of the formalin fixation,the paraffin imbedding,5μm slice.Observe the synovium apoptosis cell.The other sheets were deal with immunohistochemical staining.Observe them by microscope,choose ten eye-reach(400X) using the eye glass graticule micrometer,do the positive cell counting in every 100cell eye-reach,express them by general average and standard deviation.ResultsThe Immunohistochemical expression of MMP-7,MMP-9,TIMP-3 in the osteoarthritis patients' synovium of knee joint is more higher than the expression in the normal synovium.They are obviously difference(p<0.05).Histology consequenceFrom the result display,we can see that the normal synovial tissue of joint don't show the chronic inflammation reaction,the synovial cell distribute regulately,they don't obviously hyperplasy.The micrangium constit and the amph-tissue don't have obviously patho-change.But the OA knee synovium of joint show the chronic inflammation reaction,infiltrated a lot of achroacyte and histoleucocyte distributed by blood vessel.The synovial cell have the accrementition.The accremention show the corpora mammillaria and the fluffiness.The blood vessel hyperplasia is obvious.The monolayer blood vessel endothelium cell cellular swelling,lumen of blood vessel narrowing,the peripheral vessels fibrosis,hyalinization,chondrometaplasia.The immunohistochemical resultThe MMP-7,MMP-9,TIMP-3 are high positive express in the synovial cell,the histoleucocyte,achroacyte,the vassular endothelial ande the metaplastic cartilage.The endochylema are buffy.And in the normal synovium tissue they also have a little express.But the total statistics consequence show that the positive cell rate is obviously different with the control cell rate,P<0.05.ConclusionThe abnormal expression of MMP-7,MMP-9,TIMP-3 in the in osteoarthritis patients' synovium of knee joint was likely to be the more important index of diagnosis. |
