IntroductionThe generation and development of tumor is a complicated mechanism.Many research dedicated the biological behavior of tumor are the result of change in single or multitude gene.The activation of oncogene and deactivation of antioncoge contribute to the change from normal cells to cancer cells.The study of molecular genetics of tumor not only contribute to the diagnosis and prediction of prognosis,but also show us more clearly the pathogenesy of tumor.Though the study of molecular genetics of tumor is just in initial phase,it already give us some inspiration.The generation and development of bladder cancer are the result of change of cell molecular biology.So far,there are many report of correlation between occurrence anc progress of bladder cancer and single protein of apoptosis gene and oncogen or anti-oncogene.However there are little research about expressive sense and interrelation of several genes molecular products in the same species of transtitional cell cancer of bladder.As new members of anti-oncogene PRDMS famlily,PRDM1,PRDM2,PRDM5,PRDM14 have the typical characterize,rencent studies have show that PRDMS were expressed in many tumors.This study aims to search for the differentially-expressed of PRDM1, PRDM2,PRDM5 and PRDM14 between normal bladder cell and transitional cell carcinoma.ObjectiveTo investigate the expression and significance of PRDM1,PRDM2,PRDM5 and PRDM14 in transitional cell carcinoma of bladder(BTCC),and the expression of PRDM1,PRDM2,PRDM5 and PRDM14 were compared with the pathological grade and clinical stage.Methods Immunohistochemical(IHC) method was used in detecting PRDM1,PRDM2, PRDM5 and PRDM14 in 40 transitional cell carcinoma of bladder(BTCC) and 9 normal mucous membranes of urinary bladder which were more than 2.5cm far away from the tumors and were pathologically confirmed after cystechtomy.Then devide the 40 BTCC specimens into group by pathological grade and clinical stage,usingχ~2.test to compare the differences of each group.Results1.In 9 normal mucous membranes of urinary bladder there were no obvious positive expression.In 40 BTCC tissues,the positive rates of PRDM1,PRDM2,PRDM 5and PRDM14 were 60%(24/40),57.5%(23/40),67.5%(27/40) and 52.5%(21/40) respectively.There were significant differences of each group.2.The expression of PRDM1,PRDM14 have significant positive correlation with tumor grade(P<0.05).Expression of PRDM2,PRDM5 have significant negative correlation with tumor grade(P<0.05).3.The expression of PRDM1,PRDM14 have significant positive correlation with clinical Staging(P<0.05).Expression of PRDM2,PRDM5 have significant negative correlation with clinical Staging(P<0.05).4.The expression of PRDM1,PRDM2 have significant negative correlation (r=-0.537 ).The expression of PRDM5,PRDM14 have significant negative correlation(r=-0.446).The expression of PRDM2,PRDM5 have significant positive correlation(r=0.375).Conclusion1.PRDM2,PRDM5 overexpressed in BTCC.They linked to low histological grade,low stage and probably better prognosis.2.PRDM1,PRDM14 overexpressed in BTCC.They linked to high histological grade,high stage and probably poor prognosis.3.It may has important theoretical significance to approach the occurrence and development even prognosis of BTCC when combining to examine the expression of PRDM1,PRDM2,PRDM5 and PRDM14.
