ObjectiveTo detect the expression of VEGF-C,PI3Kp85,NF-κBp65 and Survivin in nasopharyngeal carcinoma(NPC) and chronic nasopharyngitis and to explore their role in the development of NPC as well as their interrelationships.MethodImmunohistochemical (streptavidin-perosidase SP) method was used to analyze the protein expression of VEGF-C,PI3Kp85,NF-κBp65 and Survivin in paraffin embedded tissues from 57 cases nasopharyngeal carcinoma and 20 cases chronic nasopharyngitis. Use Image-ProPlus software to determine the average optical density (MOD) of immunohistochemical Image. Then use SPSS13.0 which is statistical analysis software to analyze their relation with the clinic pathological characteristics of NPC as well as their dependablity.ResultsThe positive rate of VEGF-C,PI3Kp85,NF-κBp65 and Survivin protein in nasopharyngeal carcinoma tissues were 73.68%,78.95%,61.40% and 68.42% respectively, however those in chronic nasopharyngitis tissue were not evident, the differences were significant between nasopharyngeal carcinoma tissues and chronic nasopharyngitis tissue (P<0.05). The expression of VEGF-C and NF-κBp65 were closely related to lymph node metastasis and clinic pathological stage (P<0.05);the expression of Survivin were closely related to lymph node metastasis,clinic pathological stage and the size of primarily focus (P<0.05);but PI3Kp85 has no relation with them (P>0.05);the four factors have no relationship with sex and age (P>0.05). In addition, NF-κBp65 protein expression had positive correlation with both VEGF-C,PI3Kp85 and Survivin (r=0.413,r=0.376,r=0.526,P<0.05), and the expression of VEGF-C was positively correlated with that of Survivin(r=0.352,P<0.05).Conclusion1,VEGF-C,NF-κBp65 and Survivin were overexpressed in nasopharyngeal carcinoma tissues,and all of them have relation with lymph node metastasis and clinic pathological stage (P<0.05),So detection of them will be helpful to judge biology characteristic and metastasize potentiality of nasopharyngeal carcinoma.2,The expression of Surivivin has tumor tissue-specific and close relationship with tumor occurrence and development, so it may become an identification marker to early diagnosis and a new locus to targeting therapy.3,NF-κBp65 expression in the nucleus is a sign of its activation, its expression level in the nucleus is closely related to the development of nasopharyngeal carcinoma. Detecting the expression level of NF-κB p65 is important to judge progression of disease.4,NF-κBp65 protein expression had positive correlation with both VEGF-C,PI3Kp85 and Survivin,suggesting that they have synergistic effect in the development of nasopharyngeal carcinoma; PI3Kp85 contributes to the pathogenesis of nasopharyngeal carcinoma and lymph node metastasis may through NF-κBp65 to up regulating the expression of VEGF-C and Survivin and effect procreation and apoptosis of tumor cell.
