Objectives: The microRNAs are some small non-coding transcripts of about 22nt nucleotides which play important roles in developmental regulation. The aims of this study are to find some mouse neural development related microRNAs, and to explore molecular mechanism of neurogenesis and provide theory basis and potential treatment target on spinal cord injury.Methods : E8.5d and E9.5d mouse neural tube were selected, the expression of Nestin were detected with immunohistofluorescence and Nestin positive cells were counted by Flow Cytometry(FCM). microRNAs expression profiling in E8.5 mouse neural tube was analyzed by microarray method. Then the target genes of all these microRNAs were predicted by bio-information method (http://pictar.bio.nyu.edu, http://www.targetscan.org).Results: Almost all the neuroepithelial cells in E8.5d mouse neural tube were Nestin positive, while few cells in E9.5d mouse neural tube expressed Nestin; By FCM, the Nestin positive cells were 58.2±3% in E8.5d and 7.58±3% in E9.5d neural tube separately. Microarray analysis showed that there were 74 up-regulated and 65 down-regulated microRNAs. Some neural development related genes do have target site of the microRNAs (miR-367,miR-26b,miR-10b,miR-219,miR-199* and so on).Conclusions: Although the exactly function of the microRNAs need further study, this data implicated that microRNAs might play an important role in neural development in vivo.
