Partâ… Growth of neural stem cells in serum-free conditional medium and induce them to differentiateObjective: To investigate of culturing neural stem cells(NSCs) from rat embryos in vitro and observe their growth and differentiation.Methods: NSCs were isolated from hippocampus of SD rat embryos(P16-P18) and cultured in DMEM/F12 medium containing EGF,bFGF,B27.To observe process of cell proliferation by microscope and identify cell types by immunocytochemical analyses after differentiation.Results: NSCs grew well in serum-free conditional medium and their cell bodies present transparent with good refraction at about eighth day. After differentiation, the cells demonstrated NSE and GFAP immunoreactive.Conclusion: NSCs were cultured well in serum-free conditional medium and they could be induced to differentiate into neurons and astrocytes in serum conditional medium. Partâ…¡The study on neural stem cells differentiating into hair-like cells when co-cultured with supernatant of basal lamina from rat embryos cultured in vitroObjective: To study inducing neural stem cells(NSCs) to differentiate into hair-like cells in vitro.Methods: NSCs were isolated from brain of SD rat embryos and cultured in serum-free conditional medium. The NSCs were added to rat tail collage coated coverglasses in 6-well culture plates in DMEM/F12 medium containing 10% fetal bovine serum. The basal lamina was isolated from P0 to P3 rats and cultured in the same medium. The supernatant was added to the NSCs suspension mentioned above and co-cultured for 14-21 days. Then expression of hair cell markers( myosin VIIa and calretinin ) was detected by immunocytochemical analyses and the differetiation rate was calcuated.Results: After differentiation,15.4% of NSCs demonstrated myosin VIIa immunoreactive and 21.7% demonstrated calretinin immunoreactive.Conclusion: NSCs could be induced to differentiate into hair-like cells when co-cultured with supernatant of the basal lamina from rat embryos in vitro.Partâ…¢The study on neural stem cells differentiating into hair-like cells when co-cultured with basal lamina from newborn ratsObjective: To study inducing neural stem cells (NSCs) to differentiate into hair-like cells in vitro.Methods: NSCs were isolated from brain of SD rat embryos and cultured in serum-free conditional medium. The basal lamina was isolated from P0 to P3 rats and cultured in DMEM/F12 medium containing 10% fetal bovine serum. Then NSCs were co-cultured with basal lamina for 14-21 days. The expression of hair cell markers( myosin VIIa and calretinin ) was detected by immunocytochemical analyses.Results: After differentiation,17.1% of NSCs demonstrated myosin VIIa immunoreactive and 27.4% demonstrated calretinin immunoreactive.Conclusion: NSCs could be induced to differentiate into hair-like cells when co-cultured with basal lamina from new-born rats.
