A kind of two phase system consisting of hydrophilic organic solvent, salt and water is a novel separation system, which has some advantages, such as simple components, cheap and low-toxic raw materials, non-emulsification and low power consumption. The system can be applied to sample enrichment and separation in the fields of biochemical engineering and analytical chemistry. It is a separation system with potential industrial application.The thesis studied acetone/NaCl system and acetonitrile/NaCl system. The two-phase diagrams of these two systems were made. The partition of some semi-synthetic cephalosporins in acetone/NaCl system and acetonitrile/NaCl system was investigated. The partition of papain in acetonitrile/NaCl system was also investigated. The thesis studied the effects of concentration of separated materials, concentration of salt, concentration of organic solvents, temperature and pH on the partition in these systems. The results showed that it is feasible to separate semi-synthetic cephalosporins with these systems. The target cephalosporins can be gathered in upper phase or lower phase by adjusting pH. During the separation of papain, the enzyme and the protein could be separated at higher temperature or lower pH, but it has not application value because of high destruction rate of enzyme.The preliminary research on the separation of small molecule and macromolecule materials by the hydrophilic organic solvent/salt two phase system proved that the system has the potential to become a new separation and purification method.
