The Influence Of Different Rreatment Methods On Stored Urine Of Rabbit Ileal Neobladder Surgery Line To Intestinal Mucosa

Objective: To observe the different methods of treatment to remove rabbit intestinal mucosa and to explore a new way to remove mucosa.Methods: We divided 60 rabbits into 5 groups randomly each contained 12, and dissociated jejunum for 15 centimeteres, retained dissociated sect in order to keep blood supply, restored anastomosis for intestinal continuity. The group of A perfused 0. 9% NaCl for 10 minutes, The group of B perfused 0.5% iodophor for 10 minutes, The group of C perfused ethanol for 15 minutes, The group of D perfused 5% AgNO3 for 10 minutes.The perfusion of each group contained 20ml , The group of E had submucosal injection of distilled water, After finishing the treatment of each group we closed the abdomen layer by layer and placed drainage tube by which we perfused artificial urine for 15ml each day ,we drawed it 5 hours later . Specimen collection and indicators Detection must be done from surgery date , we collected artificial urine of each group out, centrifuged and the extracted supernatant liquid,mesurated protein content of mucus using Coomassie brilliant blue. We had dissociated intestine of each group separately in the intraoperative day , 3 days and 7 days after postoperative day, fixed the specimen using10% formaldehyde buffering and glutaraldehyde , had HE and AB-PAS staining ,oberseved specimens by electron microscope.Results: The number of goblet cells are significantly reduced when the treatment group compared with the control group under the per unit area (P <0.005), we can see obvious chemical injury in mucous layer of each treatment group by visible light microscopy, goblet cell damaging, cell injury and gland cell apoptosis under electron microscope, the group of submucosal injection of distilled water in particular.Conclusion: The injection of Iodophor, ethanol, 5% AgNO3 solution and distilled water all have the effect of removed intestinal mucosa , the intestine sect of submucosal injection of distilled water can be completely removed, no significant fibrosis, no significant intestinal contracture, having important clinical meaning for ileal neobladder.
