Objective The purpose of this study is to explore the developed mecha- nism of dilated cardiomyopathy (DCM) by investigating the changes of CD4 ~+ CD25 ~+ T cells and CD4 ~+ CD25 ~+ Foxp3~+ T regulatory cells in peripheral blood mononuclear cells from patients with dilated cardiomyopathy .Methods Thirty patients (19 males and 11 females) with dilated cardiomyopathy were collected into the study and twenty healthy individuals (12 males and 8 females) served as controls. 2 ml of venous blood from DCM patients or from control subjects were collected, anti-agglutinated with EDTA, Peripheral blood mononuclear cells(PBMCs) were obtained by lymphocyte's separating medium and stained by monovlonsl antibody; the alone expression or combined expression of CD4,CD25,Foxp3 in lymphocyte were examined by flow cytometer.Results There were no significant difference between DCM patients and healthy individuals as to the expression of CD4 on the lymphocyte(P > 0.05). However, compared with health control, low level CD25 Foxp3 was expressed in the CD4~+ T cell(P <0.05) and lower level CD25 was also expression on the CD4~+ T cell (P <0.01).The frequencies of Treg was markedly lower in patients with bad heart function than those of in patients with good heart function .Conclusions The frequencies of CD4~+CD25~+ Treg decrease in patients with DCM and are associated with heart function. The reduce of CD4~+CD25~+ Treg may lead to the function of inhibit- autoimmune down regulation and bring about autoimmunity cardiomyopathy.
