| Objective:To examine the expression of TGF-a,HIF-1a,VEGF in the issues with pancreatic adenocarcinoma and normal pancreas by the immunohistochemisty method and study the changes and the relationship of these proteins expression in pancreatic adenocarcinoma.Methods:The expressions of the TGF-a,HIF-1a,VEGF were examined in formalin-fixed paraffin embedded samples of 41 patients with pancreatic adenocarcinoma and 12 normal pancreatic issues by SABC immunohistochemical method.The relationships between the expression and clinical characters were analysied, such as,ages,sex,size of the tumor,location of the tumor,pathological grade,TNM staging and the lymphatic metastasis.Results:(1)The positive rate of TGF-a was 70.73%(29/41) in the pancreatic adenocarcinoma,positive staining was mainly located in the cytoplasm and/or on the surface of pancreatic carcinoma cells.In the normal pancreatic tissue,the positive rate of TGF-a was 16.70%(2/12).Between the pancreatic carcinoma group and the normal pancreatic tissue group,the positive rate of TGF-a was significant difference(X~2= 90064,P<0.01),and there were no significance about the age,sex,size of the tumor, location of the tumor,pathological grade,TNM staging and the lymphatic metastasis. (2)Positive staining of HIF-1a was mainly located in the cytoplasm and/or in the nucleus of pancreatic carcinoma cells with the positive rate of 63.41%(26/41).In the normal pancreatic tissue,the positive rate of HIF-1a was 0%(0/12).There were significant differences(X~2=14.938,P<0.01) when comparison was made between pancreatic carcinoma group and normal pancreatic tissue group,and there were significance between the comparisions of size of tumor,staging of TNM and the lymphatic metastasis. (3) VEGF was stained mainly in the cytoplasm and/or on the surface of pancreatic carcinoma cells with positive rate of 76.61%(31/41).In the normal pancreatic tissue,the positive rate of VEGF was 0%(0/12).There were significant differences(X~2=19.826, P<0.01) when comparison was made between pancreatic carcinoma group and normal pancreatic tissue group,and there were significance between the comparisions of size of tumor,staging of TNM and the lymphatic metastasis.(4) A positive correlation was observed between the expression of HIF-1a and VEGF in pancreatic carcinoma(r=0.474, P<0.05).Conclusions:The overexpression of TGF-a,HIF-1a and VEGF were found in pancreatic adenocarcinoma.The HIF-1a.may induce the angiogenesis in pancreatic adenocarcinoma by upregulating the transcription of VEGF gene.These are important in the occurrence,progression and metastasis of the pancreas cancer. |
