Objective Epstein—Barr virus(EBV) is an ubiquitous sherpes virus which is the causative agent of infectious mononucleosis,EBV gene has been identified in about 10%of gastric carcinomas.EBVaGC is rich in lymphoid-cell infiltration and appears to have a better prognosis than EBV-negative carcinoma.In order to explore the Potential Pathogenic role of protein VEGF,CD34,CD44v6 in EBVaGC,We deteeted the expression of VEGF,CD34,CD44v6 in EBV—assoeiated gastric carcinoma(EBVaGC),compared some EBV—encoded proteins in EBVaGCand clinicopathological data matched EBV—negative gastric carcinoma(EBVnGC).Methods The expression of VEGF,CD34,CD44v6 Protein were tested by imimmunohistochemistry in 15 EBVaGCs and 45 EBVnGCs.RT-PCR and Southern hybridization were used to detect the expression of nuclear antigen(EBNA) EBNA 1 and 2, latent membrane protein LMP1,and lytic genes(early genes BARF1 and BHRF1) in EBVaGCs.Results The positive rate of VEGF expression in 60 cases with gastric carcinoma was 71.67%(43/60) while that of the adjacent tissue was 88.34%(53/60),There was obvious difference between the two groups(P=0.022).MVD of gastric carcinoma was obviously lower than that of adjacent tissue(P=0.000),The positive rate of CD44v6 expression in gastric carcinoma was 61.67%(34/60),and that of the adjacent tissue was 48.34%(29/60). There was no difference between the two groups(P=0.142).There is significant correlation between the expressions of VEGF,MVD,CD44v6 and TNM stage,depth of infiltration and lymph node metastasis(P<0.05).The positive rate of VEGF expression in EBVaGCs was 80%(12/15),and that of the adjacent tissue was 68.9%(31/45).There was obvious difference between the two groups(x~2=3.928,P=0.047);The MVD in EBVaGCs(44.60±14.43) was lower than that of EBVnGC(46.11±17.80),while There was no difference between the two groups(P=0.946).The positive rate of CD44v6 expression in EBVaGCs was 66.7%(10/15),and that of the adjacent tissue was60%(27/45).There was obvious difference between the two groups(x~2=8.349,P=0.039)Conclusions The expressions of CD44v6,CD34 and VEGF are correlated to the invasion and metastasis of gastric carcinoma.Detection of VEGF,MVD and CD44v6 expression in gastric carcinoma is helpful to evaluate the infiltration,metastasis and prognosis of gastric carcinoma.The over expression of VEGF and CD44v6 in EBVaGC don't play a role in the vascularization and lymph node metastasis,so EBVaGC appears to have a better prognosis than EBV-negative carcinoma. |
