Objective:To investigate the relationship of serum Adiponectin (ADPN),Leptin(LEP) concentration, adiponectin gene(APM-1)single nucleotide polymerphisms (SNPs) and breast cancer incidence,and to find out the pathogenesis of breast cancer.Methods:A case-control study was used.There was 68 primary female breast cancer patients who were cured in tumor hospital of Shanxi from 2008-04 to 2008-10 in case,and 62 healthy people came from the same place in control. 2ml non-anticoagulant blood and 2ml anticoagulant blood from elbow vein of investigation object were collected.Blood serum was isolated from non-anticoagulant blood,and the levels of serum adiponectin and leptin were measured by Enzyme-Linked Immunosorbent Assay(ELISA);The DNA was extracted from anticoagulant blood,the single nucleotide polymorphism(SNP) of APM-1 gene T45G was detected by Polymerase Chain Reaction Restriction Fragment Length Polymorphism Analysis (PCR-RFLP).All objects were investigated including height,weight,waistline,breechline,and all were calculated the body mass index (BMI),waist-to-hipratio(WHR).Results:①The serum ADPN concentration(5.98±1.96)μg/ml in breast cancer group was significantly lower than in the healthy control(10.78±3.36)μg/ml(p<0.01); the serum LEP concentration(15.48±6.52)ng/ml in breast cancer group was significantly higher than in the healthy control(12.52±6.09)ng/ml(p<0.01);and there were significant differences of the serum ADPN and LEP between postmenopausal and premenopausal women with breast cancer(p<0.01).②There was a protective correlation between ADPN and breast cancer incidence(OR=0.34,95%CI:0.25~0.46).③There was negative correlation between ADPN level and LEP,BMI,WHR,its correlation coefficient was 0.84, 0.40, 0.58, respectively. All p< 0.01.④TT,TG and GG genetic frequency of APM-1 gene SNP+45 in the breast cancer group were 45.59%,41.17%,13.24%,respectively. allelomorphic gene T and G genetic frequency were 66.17%and33.83%,compared with healthy control group TT 53.22%,TG 38.71% and GG 8.06%,allelomorphic gene T 72.58% and G 27.42%,there were no significant differences (p>0.05).Conclusion:①Serum lower ADPN and higher LEP levels had significant correlations with breast cancer.②Dereased serum ADPN levels may be a risk factor of breast cancer.③APM-1gene SNP+45had no significant correlation with breast cancer.
