| Objective Discussion of hepatocyte growth factor(HGF) on the high glucose environment, human proximal tubular epithelial cells(HK-2) proliferation and TGF-β1 expression in order to reveal the HGF in diabetic nephropathy(DN) in the protection of renal tubular epithelial cells Mechanism of action.Methods The experiment is divided into the control group,high glucose group,high glucose + HGF group,renal tubular epithelial cells in vitro(HK-2),with MTT testing for more than three sets of cells were incubated 12h,24h,48h,72h,96h cell vitality.Enzyme-linked immunosorbent assay(ELISA) testing for more than three groups of cells in the supernatant 12h,24h,48,72h,96h transforming growth factorβ1(TGF-β1) concentration.Results①High glucose HGF joined the group after 12h,24h,48h detection of cell viability high glucose group had no significant changes in the((P> 0.05),72h,96h high glucose testing cell viability was significantly higher(P <0.01);with the control group no significant changes in cell viability(P> 0.05).②After 24h,stimulated by high glucose concentrations of TGF-β1compared with the control group increased significantly(P <0.01);After added HGF into the high culture,and high glucose group compared with the concentration of TGF-β1 significantly lower(P<0.01),and no significant difference was found in control group(P>0.05).Conclusion HGF can inhibit HK-2 in the release of TGF-β1 and the protection of high glucose-induced tubular epithelial cells. |
