Objective: To investigate the brain protective effect and possible mechanism of Vitis quinquangularis Rehd (VRE)through the establishment of Cerebral Ischemia reperfusion model. Provide experimental evidence to further research and development the drug effects of VRE.Methods:(1) Experimental grouping and drug delivery :SD rats were randomly divided into six group: Normal group, Control group, Nimodipine group ,VRE large-dose group, VRE medium-dose group, VRE low-dose group. The rats were equally divided between male and female and each group ten. Their weight were(250±20)g. the next day they were gavaged administration for seven days. the dose was Nimodipine group 5mg·kg-1·d-1 ,VRE large-dose group 4g·kg-1·d-1, VRE medium-dose group 2g·kg-1·d-1, VRE low-dose group 1g·kg-1·d-1.meanwhile Normal group and Control group with Saline. (2) The establishment of cerebral ischemia reperfusion model: Refered to longa suture ways establish the middle cerebral artery occlusion MCAO model。After ischemic for two hours, Removaled the suture and reperfusion for twenty-two hours.then carried out nerve function evaluation first And detected the content change of LDH,NO,MDA in plasma. Brains were averagly divided into five pieces from amount of very to occipital. About 1.8mm one piece. the brains were stained in 4% TTC with 37℃f or 30 minutes, then they were fixed in 4% Polyformaldehyde for 24 hours . The photograph ischemic area were statisticsed with AutoCAD Soft ware.We used immunocytochemical method detecte the expression of bcl-2 and caspase-3. Results:(1) The effect of VRE on nerve function and ischemic area. Drug-treated group could improve the nerve function situation and lightened the brain ischemic area. ( p<0.05 vs control group) the drug effects of VRE medium-dose group and VRE low-dose group were better than Nimodipine group and VRE large-dose group.(2)The effect of VRE on the expression of bcl-2 and caspase-3. the expression of bcl-2 and caspase-3 were seen at ischemic surrounding area .But the expression of bcl-2 was least and caspase-3 is most in control group. VRE groups could increase the expression of bcl-2 and decrease the expression of caspase-3 ( p<0.05) vs control group) ,especially the VRE medium-dose group was best.(3) The effect of VRE on the level of LDH,NO,MDA. VRE groups could decrease the level of LDH,NO,MDA in plasma ( p<0.05 vs control group), especially the VRE medium-dose group was better than other groupsConclusion: (1) VRE could protect brain injure with cerebral ischemia-reperfusion , That was VRE could improve the nerve function situation and lighten the brain ischemic area . (2) VRE could obviously decrease the level of LDH,NO,MDA in plasma and increase the expression of bcl-2 and decrease the expression of caspase-3 at ischemic surrounding area. Those changes relevanted to the VRE protected brain injure with cerebral ischemia-reperfusion(3) The results of this article illustrated the role of VRE protect brain injure with cerebral ischemia-reperfusion was better than Nimodipin , which illustrate VRE had Development prospects and Practical value.
