【Objective】To explore the effect of Gastrodin and Total Saponin of Panax Notoginseng on the expressional changes of ten ageing-associated genes in the hippocampus and frontal lobe of SAM/P-8.【Methods】The expressional changes of aging-associated gene mRNA in hippocampus and cortex of frontal lobe of SAM/P-8 were analyzed by RT-PCR.In addition,observes the SAM/P-8's behavior study change through the Morris water maze.【Results】(1) All of the aging-associated gene were detected in hippocampus and frontal lobe of SAM/P-8 in apply Gastrodin and Total Saponin of Panax Notoginseng,but level respectively different.(2) The expression of SCN2B mRNA in hippocampus of adult mouse increased following Gastrodin administration;MAP1B mRNA and RAP2A mRNA expression increased in cortex of frontal lobe,while the expression of Sortilin-1 mRNA decreased in hippocampus.(3) Compared with aging control,the expression of MAPKK4,Sortilin-1 and Rab6A mRNA in Gastrodin treated group should decrease.(4) The expression of SCN2B gene increased significantly compared between adult PNS-treated group and adult control group.(5) The expression of MAP2,Sortilin-1,Rab6A,Calcineurin-B and MAPKK4 genes decreased compared between the senium PNS-treated group and senium control group.(6) There was no expressional changes of MAP1B,Calmodulin-1,Regucalcin and RAP2A between every groups.【Conclusions】(1) All of the aging-associated genes might play important roles in aging.(2) Treatment with Gastrodin and Total Saponin of Panax Notoginseng could regulate the gene expressions of SCN2B,MAP1B, RAP2A,which made the anti-aging effects in the hippocampus and the frontal lobe of adlut SAM/P-8.(3) Gastrodin or Total Saponin of Panax Notoginseng might make the anti-aging effects in old SAM/P-8 by the methods of expressional regulation in some genes(including MAPKK4,RAB6A,Sortilin-1,Calmodulin-1 and MAP2),especially Sortilin-1 and MAPKK4.(4) The anti-aging effects of Total Saponin of Panax Notoginseng in frontal lobe of SAM/P-8 might not be refered the 10 genes detected in the present work..(5) Total Saponin of Panax Notoginseng and Gastrodin might take the different roles in anti-aging by reversing the gene expressions of Calmodulin-1 and Regucalcin,in the hippocampus and the frontal lobe of SAM/P-8.
