Objective To investigate the effect of anisodamine on expression of HSP70 and Caspase-3 in hippocampus CA1 region of rats with burn injury,and discuss the probable mechanism and protective of anisodamine for the rats brain tissue after burn.Methods We took 45 healthy SD rats(provided by kunming Medical college)to set up burned models,which were randomly divided into control group,burned group and treatment group.We took the brain tissue sample separately after 6,12,24,48 hour post burn injury.In the treatment group,Ani 15mg·kg-1,ip after burn injury,the expression of HSP70 and Caspase-3 in the hippocampus CA1 region was detected with immunohistochemical methods and analyzed by a micro-image analysis system,and at the same time we observed the pathological changes of the brain tissue of each groups.Results In the burned group,the expression of HSP70 was higher than the one in control group at equal and parallel time(P<0.05),6-24H post burn injury the expression was increased,while the expression of HSP70 decreased obviously 48H post burn injury.In the treatment group,the expression was higher than the one in the burns group (P<0.05),at equal and parallel time.In the burned group,the expression of Caspase3 was higher than the one in control group at equal and parallel time(P<0.05) 6-24H post burn injury the expression was increased,while the expression of Caspase3 decreased obviously 48H post burn injury.In the treatment group,the expression was lower than the one in the burns group at equal and parallel time(P<0.05).In pathological observation,the control group manifested the intact brain tissue formation,the burned group showed obvious brain tissue injury in the early phase of burn injury,while the treatment group showed much less severe injury than the burned group.Conclusions In the early phase of severe burn injury,the brain tissue injury is very severe.It may be one of the vital mechanisms of anisodamine protecting brain tissue,which anisodamine increases the expression of HSP70 and decreases the expression of Caspase-3 in the brain tissue of SD rats in the phase of burn injury.
