| Objebtive This study was to evaluate the effect of Janus kinase inhibitor AG490 and signal transducer and activator of transcription(STAT) signaling pathway in the pathogenesis of TNBS - induced experimental colitis in rats.Methods TNBS-induced experimental colitis was constructed in rats. Rapamycin(RPM)and AG490 treat rats.Pathological score was used for evaluating the colonic inflammation.In addition,Western blot method were used to detect the expressed level of MMP-1,MMP-2,MMP-3 and ITMP-1.Gelatinases activities of the colitis were analyzed by sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS - PAGE) gelatin zymography.Results AG490 treatment group exhibited lower pathological score:5.5±2.16.That of RPM treatment group is:5.17±1.80(P<0.05).The expressed level of MMP-1,MMP-2 protein in colonic tissue of AG490 group and RPM group were both significantly lower than control group.There were no diference in MMP-3 and TIMP-1 at protein level among AG490 group,RPM group and control group.Activity of MMP-2 was obviously reduced both in AG490 group and RPM group,as compared with that in the control group. |
