【Objective】:To establish the model of pure cerebral concussion(PCC) and multiple cerebral concussion(MCC) in rats,observe the changes of the brain derived neurotrophic factor(BDNF) in the different brain regions of PCC and MCC and the changes of neurite growth inhibitor-A(Nogo-A) in the different brain regions of PCC and MCC and the changes of BDNF and Nogo-A in the different brain regions of PCC and the changes of BDNF and Nogo-A in the different brain regions of MCC so as to understand the neurobiologic mechanisms of cognitive behavioral dysfunction after PCC and MCC in rats.【Methods】:A rat PCC model and a MCC model was duplicated respectively by using a metallic pendulum -striker concussive device.In Each model have forty-eight male Sprague-Dawley rats which were randomly divided into six MCC groups:1d,2d,4d,8d,16d and 24d group(n=6,in each group).One control was used(n=6).The BDNF and Nogo-A of the different encephalic region which include septal area section,hippocampus section,brainstem section and so on were stained with anti-BDNF antibody and Anti-Nogo-A antibody so as to detectde by the ambi-immunohistochemistry S-P method.【Results】:(1)The optical density value results of the BDNF in the different brain regions of PCC and MCC as followed:The positive optical density value of BDNF after PCC increased gradually and reach a peak in the prophase of post-injury.Compared with the control group,the positive optical density value of BDNF of secondary motor cortex(M2) in the 1d group and of lateral septal nucleus, intermediate part(LSI) in the 2d group and of accumbens nucleus(AcbC) in the 2d group and of Tu in the 2d group and of retrosplenial agranular cortex(RAS) in the 2d group following PCC,reached a peak,there were statistical significance(p<0.05).Due to a transitory decrease in the 1d groupe following MCC and increase grandually,the time reach a peak of positive optical density value of BDNF would be later following MCC than PCC,but last more long.The positive optical density value of BDNF of field CA1 of hippocampus -2(CA1-2) decrease in the 1d group and to a peak in the 4d groupe,of field CA3 of hippocampus(CA3) decrease in the 2d group and to a peak in the 8d groupe,the difference of the value above all there were statistical significance(p<0.05) compared with the control group.Compare with the control group the optical density value of BDNF of Den which reach a peak in the 24d group,there were statistical significance(p<0.05) in the 4d and 8d and 16d and 24d group.(2) The optical density value results of the Nogo-A in the different brain regions of PCC and MCC as followed:to a peak following PCC and decreace soon after,for example reach a peak in the 1d group following PCC in M2 revealed statistical differences compared with the control group(p<0.05),decrease soon after, almost recovered in the 4d group.Reach a peak in the 1d group following PCC in Cpu-B revealed statistical differences compared with the control group (p<0.05),decreased soon after,almost recovered in the 4d group.The time reach peak to be later following MCC than PCC,but the extent to be more larger and last more longer compare with PCC,for example reach a peak in the 24d group following MCC in caudate putarnen(Cpu-B),and revealed statistical differences compared with the control group(p<0.05) in the 2d,4d,8d,16d,24d group.(3) The optical density value results of the changes of BDNF and Nogo-A in the different brain regions of PCC as followed:the optical density value results of the positive reactant of the Nogo-A increase in the prophase,reach a peak in the 1d or 2d group in the encephalic regions where were bet hardly following PCC,decrease soon after;the optical density value results of the positive reactant of the BDNF increaseing when the Nogo-A decreasing,reach a peak in the 4d and 8d group,for example in the encephalic regions of LSI and Cpu-B.(4) The optical density value results of the changes of Nogo-A and BDNF in the different brain regions of MCC as followed:the optical density value results of the positive reactant of the Nogo-A increase and maintain in a high level,reach a peak in the 8d group,for example in the encephalic regions of primary somatosensory cortex,barrel field(SIBF) which revealed statistical differences compared with the control group(p<0.05).The optical density value results of the positive reactant of BDNF reach the first peak befor the peak of the Nogo-A and to reach the second peak subsequently,for example the optical density value results of the positive reactant of BDNF reach the first peak in the 4d group and reach the second peak in the 16 group.【Conclusions】:1.The pure cerebral concussion(PCC) and multiple cerebral concussion(MCC) model in rats was established successfully using a metallic pendulum-striker concussive device;2.The expression of the positive reactant of the BDNF increase in different level following PCC,especially serious on the 4th day post-injury,and increase in different level following MCC.,especially serious on the 8th day post-injury,for example in the encephalic regions of M2,Cpu-LV,CA1-3,CA2,SIBF,Pno and so on.3.The expression of the positive reactant of the Nogo-A increase in different level following PCC and MCC respectively,especially serious on the 1th day and 2th day post-injury following PCC and on the 4th day and 8th day post-injury following MCC,it is later the time reach a peak of the value following MCC than PCC but the extent to be more larger and last more longer,for example in the encephalic regions of M2,primary somatosensory cortex,upper lip region (S1Ulp),olfactory tubercle(Tu),CA2,SIBF,pontine reticular nucleus,oral part (Pno) and so on.4.The expression of the the positive reactant of the Nogo-A to reach a pesk earlier than the positive reactant of the BDNF following MCC,for example in the encephalic regions of M2,Cpu-LV and LSI.5.The optical density value results of the positive reactant of the Nogo-A increase and maintain in a high level,reach a peak in the 8d group,the optical density value results of the positive reactant of BDNF reach the first peak befor the peak of the Nogo-A and reach the second peak subsequently,for example in the encephalic regions of SIBF and CA2 and so on.
