| [Objective]To study the transdifferentiation changes on the immortalized rat alveolar cells typeâ…¡,which induced by the soot of Yurman Tin smelting works in vitro,and to give some experimental basis for the etiology and pathogenesis reaserch on the Yunnan tin miner lung cancer.[Methods](1) Immortalized rat alveolar cells typeâ…¡was treated by the soot of Yunnan Tin smelting works for 72 hours every other generation until the 9th generation.From the 11th generation,it was cultured by normal method until 35th generation.(2)The cell morphology was observed with inverted microscope.(3)The cell cycles were analyzed through flow cytometry.(4)The transformation of epithelial cells was identified by anchorage independent growth in soft agar.(5) The expression of CTGF,Vimentin,AQP-5,SP-C,MDM2 were detected by western blotting.[Results](1)The soot treated alveolar cells typeâ…¡changed a lot after 25th generation. The cell morphology and structure under light microscope and transmission electron microscope different from the untreated cell.the treated cell can grows in the soft agar as cell colonies,the coloning efficiency associate with the cultrue generation(p< 0.05)and not associate with the molarity of the soot(p > 0.05).The S period propotion of Cell cycles in the treated cell is higher than the untreated cell.(2)The CTGF and MDM2 immunocytochemistry results of the 25th and 30th cells were positive,and the blank was nagative.(3)The western blotting results of 25th,30th,35th cell show that there was expression of SP-C,but there were no expression of vimentin and AQP-5;the blank was opposite.[Conclusions](1)The soot of Yunnan Tin smelting works can induce malignant transformation of immortalized rat alveolar cells typeâ…¡(RLE-6TN).We have built the model of malignant transformation of immortalized rat alveolar cells typeâ…¡(RLE-6TN) induced by the soot of Yunnan Tin smelting works.(2) The soot of Yunnan Tin smelting works can induce malignant transformation of RLE-6TN cell, and the malignant transformation level is positive correlate with cell genneration after induced by the soot,but is not association with the the molarity of the soot.(3)During the transformation process,the treated cell los some features on alveolar cells typeâ…¡, and express CTGF.The milignant of the cells were associated with MDM2 high expressing and FHIT not expressing. |
