The Expression Of β-catenin And CyclinD1 In Extramammary Paget's Disease | Posted on:2010-05-02 | Degree:Master | Type:Thesis | Country:China | Candidate:J Shi | Full Text:PDF | GTID:2144360275492604 | Subject:Dermatology and Venereology | Abstract/Summary: | PDF Full Text Request | ObjectiveTo explore the expressions and the relationship between expressions of P-catenin and cyclinDl in the tissue of Extramammary Paget's disease(EMPD).Crocker,in 1889,described the first patient with extramammary Paget's disease.Extramammary Paget's disease(EMPD) is a rare malignant intraepithelial neoplasm of genital region, perineum or apocrine gland bearing skin in aged people.and is frequently associated with underlying sweat gland or visceral malignancy.It's characterized by eczematous lesions in clinical and Paget's cells in histological.The histogenesis of EMPD has long been a subject of controversy.Studies show that Wnt/β-catenin signal transduction disorder could be close correlated to cancer.Our study focus on the expression of P-catenin used S-P immunohistochemistry technology and analyze the relationship between expression ofβ-catenin and cyclinDl.So as to provide objective evidence for clinical diagnose and therapy of EMPD.MethodsS-P immunohistochemistry technology was used to detect the expression ofβ-catenin and cyclinDl protein in 32 cases of EMPD,30 cases of chronic eczema in anogenital region.Laboratory data were the analyzed statistically together with the related clinical-pathological data.ResultsThe abnormal expression rate ofβ-catenin in 32 cases of EMPD was 75% (24/32),which was significantly higher than that in chronic eczema,which abnormal expression rate is 27%(8/30),the difference was statistically significant(P<0.05). The expression ofβ-catenin was significantly correlated with dermis infiltration, lymph node metastasis in EMPD(P<0.05).The positive expression rate of cyclinDl was 91%(29/32),which was significantly higher than that in chronic eczema,which positive expression rate is 40%(12/30),the difference was statistically significant(P<0.05).The expression of cyclinDl was significantly correlated with dermis infiltration, Iymph node metastasis and in EMPD(P<0.05).The positive expression rate of cyclinDl in patients with normal and abnormal expression ofβ-catenin were 43% (13.30) and 84%(27/32) respectively.The difference was statistically significant(P<0.05).Conclusionsβ-catenin was a kind of poly functional cytoplasm protein and a key molecular of Wnt signal pathway which was closely correlated with tumorigenesis,infiltration and metastasis of tumor.The abnomal activation of Wnt-βcatenin signal pathway played an important role in tumorigenesis,so recarding Wnt-βcatenin signal pathway as the origin of oncotherapy by drug becomed a new research field.Of course,some kinds of blocking agent acting on this signal pathway have been finded and just researched,but their potential effects on oncotherapy still need be investigated and discussed.CyclinDl gene has been confirmed a kind of proto-oncogene and the regulating cellcycle gene in the most direct connection with tumor.CyclinDl was the target gene ofβ-catenin activation in cell nucleus.Whenβ-catenin accumulated in cytoplasm,it may activate CyclinDl genetic transcription.Overexpression of CyclinDl may promote cell multiplication and lead to disturbance of cellcycle.In the tissue of EMPD,ectopic expression ofβ-catenin protein activated the overexpression of CyclinDl and resulted in proliferation and disordered differentiation,soβ-catenin may play an important role in genesis of EMPD. | Keywords/Search Tags: | β-catenin, CyclinD1, Wnt signal transduction, Extramammary Paget's disease, Immunohistochemistry | PDF Full Text Request | Related items |
