Objective To investigate the expression of granulocyte colony-stimulating factor receptor(G-CSFR,CD114) in acute myeloid leukemia(AML) bone marrow CD34~+ cells and evaluate G-CSF's secutiy of applying to the patients of AML after chemotherapy.Methods 70 AML patients[37 deno-vo or relapsed AML patients,33 AML patients in complete remission(CR)]and 23 normal controls were detected for the expression of G-CSFR in CD34~+ cells by Fluorescence-activated cell sorer(FCM).62 AML patients [33 deno-vo or relapsed AML patients,29 AML patients in complete remission(CR)] and 16 normal controls were detected for the expression of G-CSFR in BMMNC by Semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR).Results:1.The ratio of CD114~+CD34~+/CD34~+ in the deno-vo or relapsed group,CR group and the control group were(11.69±14.83)%,(31.84±18.74)%and(32.87±21.56)% respectively.deno-vo or relapsed group's was lower than CR group's and the control group's(P<0.05),there was no significant difference between CR group's and the control group's(P>0.05).2.G-CSFR mRNA expression in BMNNCs of the deno-vo or relapsed group,CR group and the control group were(30.52±11.28)%,(85.13±21.25)%and (91.57±18.64)%respectively,Deno-vo or relapsed group's was lower than CR group's and the control group's(P<0.05),there was no significant difference between CR group's and the control group's(P>0.05).3.10 AML patients were followed up,The ratio of CD114~+CD34~+/CD34~+ before treatment and in CR were(12.58±14.57)%and(30.13±17.14)%respectively,The ratio before treatment was lower than that in CR(P<0.05),G-CSFR mRNA expression in BMNNCs of the patients before treatment and in CR were (32.23±13.84)%and(83.27±19.79)%respectively,there was a significant difference (P<0.05).Conclusions:1.The expression of G-CSFR in AML bone marrow CD34~+ cells was lower than that in normal CD34~+ cells,G-CSF in the treatment of AML after chemotherapy should be safe.2.The expression of G-CSFR was lower in deno-vo or relapsed group,and higher in CR group,so,the number of leukemia cells related to the lower level of G-CSFR, which means that it can be used as an index for the diagnosis and therapy response of AML. |
