PartⅠIdentification The Expression of SOCS3 on Normal and Unexplained Spontaneous Abortion DeciduasObjective:To initial observe the expression of SOCS3 on normal and Unexplained spontaneous abortion deciduas,and the results can be fundamental for how The Formula of Tonifying Kidney of Vital Essence regulating the immunity system of deciduas.Methods:Collecting the deciduas of normal and unexplained spontaneous abortion using immunohistochemistry for detecting whether there is expressing SOCS3 protein.Results:On both deciduas of normal and unexplained spontaneous abortion,the SOCS3 is found in not gland but also interstitial.Conclusion:Both the normal and unexplained spontaneous abortion deciduas can express SOCS3.PartⅡNourish Kidney Invigorating Qi Formula modulates the secretion of cytokines of Th1 and Th2 cells by CD4 positive T cells of human decidualObjective:To explore whether The Formula of Tonifying Kidney can modulate the secretion of IL-4 and IFN-γof the CD4~+T cells from human decidua during first trimester,which will lead to further investigate the anti-abortional effect mechanisms of The Formula of Tonifying Kidney.Methods:40 female adult SD rats were divided randomly into two groups:one group were gavaged with The Formula of Tonifying Kidney;the other were gavaged with Sodium Chloride serum.Two groups were gavaged with same volume for 3 days,then the blood was collected from carotid artery.Human decidua tissues were collected by artificial abortion at 6～10 weeks of gestation.The CD4~+T cells were separated by MACS.CD4~+T cells were divided into four groups during culturing.Group A was treated with 10%fetal calf serum,it is the control group;Group B was treated with 10%herbal medicine serum;Group C was treated with 20%herbal medicine serum; Group D was treated with Sodium Chloride serum.The supernatants were harvested respectively from the cultured cells after having been treated with different serums for 24,48,72h.The levels of IL-4,IL-10 and IFN-γ,TNF-αin the culture supernatants were measured by ELISA.Results:Both Group B and C can improve the level of IL-4 and IL-10 evidently (P＜0.05);at the same time they can inhibitor the IFN-γintensely(P＜0.01),but only 24 hours of group B can inhibitor the TNF-αevidently(P＜0.05).Conclusion:Nourish Kidney Invigorating Qi Formula improves the Th2 type cytokine via up-regulating IL-4,IL-10 and down-regulating IFN-γ,TNF-αof the decidual CD4~+T cells.
