Objective:(1) Observe the vital sign of tumor-bearing mice after given PAMD(phenolicalkaloids of Menispermum dauricum).(2) To observe the inhibitive effect of PAMD on the proliferation of metastatic tumor model of tumor-bearing mice(3) To observe the influence on immune organ of tumor-bearing mice.(4) To observe the effect on expression of MMP-2 of tumour tissues.(5) To observe the effect on expression of MMP-9 of tumour tissues.Methods:The basic status of mice and the metastatic tumor model of tumor-bearing mice was established with BXPC-3 cell line,the inhibition rate of tumor proliferation was detected after treatment with PAMD. Meanwhile,the effect on immune organ index was detected .Immunohistochemistry was carried out to confirm the expression of MMP-2 and MMP-9 after treatment of tumor-bearing mice with PAMD.Results:PAMD could inhibite the tumor growth of tumor-bearing mice of BXPC-3 cell line,The inhibition rate were 34.91%,52.83%,41.51%, respectively.Compared with the control group with statistical significance (P<0.01).PAMD also could not show markedly effect on basic status of mice and the immune organ index.Meanwhile.PAMD could decrease the content of MMP-2 in tumour tissues of tumor-bearing mice,compared with the model group with statistical significance(P<0.05 ).PAMD could inhibit the expression of MMP-9 remarkable(P<0.01).Conclusions:PAMD showed markedly inhibiting action on the tumor growth of tumor-bearing mice,but showed no serious adverse reactions on basic status of mice and the immune organ index.The anti-tumor mechanism of PAMD through decreasing the content of MMP-2 and MMP-9 in tumour tissues of tumor-bearing mice,which provides stable scientific basis for reasonable exploiture and use in clinical.
