| Objective:To observe the effects of recuperating-heart granule which can tonify Qi,nourish blood,stabilize the heart and tranquilize the mind on rabbit model of similar unstable angina pectoris and the expression of serum PAPP-A.To investigate its mechanism of action and to provide a objective basis for this therapy.Methods:Rabbits were given arteria carotis communis injury by using an jntravascular balloon before they are fed on a high fat diet for 8 weeks. To confirm forming atherosclerosis plaque by color ultrasonography,then they are randomly divided into model control group,recuperating-heart granule group,Cedocard group and atorvastation group.There are 7 rabbits in every group.Check another seven rabbits as blank group.Rabbits in every group are lavaged drugs or distilled water for two weeks.At the end of week 10,to copy the model of similar unstable angina,pectoris by using the method of Pharmacological triggering.To survey the content of serum PAPP-A by enzyme linked immunosorbent assay.To investigate the thickness and blood flow rate of arteria carotis communis by body surface chromatic color ultrasonography and to observe pathomorphism change of arteria carotis communis by HE dyeing.Results:1.According to the result of PAPP-A,compared with blank group, PAPP-A increase obviously in each group at the end of 8 weeks,it is considered statistically significant(P<0.01).At the end of week 10,PAPP-A decline in treatment groups,to compare with model control group,it is considered statistically significant(P<0.01).PAPP-A levels of Recuperating-heart granule group and atorvastation group are significantly lower than Cedocard group(P<0.05).But the therapeutic effect between them is similar.2.According to the results of body surface ultrasonography,every group emerge that arteria carotis communis IMT obviously thicken,PI and RI addition at the end of 8 weeks,compared with blank group(P<0.01 or P<0.05),but Vs of every group accelerate nubilously(P>0.05).At the end of week 10,IMT PI and RI of every treatment group were decreased.Compared with model control group,they are considered statistically significant(P<0.01 or P<0.05),but IMT in Cedocard group descent obsoletly and it is considered statistically significant(P>0.05).IMT of recuperating-heart granule group and atorvastation group are lower than Cedocard group(P<0.05).Difference in recuperating-heart granule group and atorvastation group is nonsense.3.Pathomorphism research indicated,besides black group,every group emerge that vascular endothelial cell damaged,endangium thicken,fatty deposition obviously,fatty substance plaque formed.Compared with model control group,each treatment group's arteria carotis communis endothelial cell decrease lightly,endomembrane thicken lightly,fatty deposition decrease under endomembrane,arterial media vascular smooth muscle cells proliferation,adventitia fibrosis and chronic inflammation cell infiltrated reduced to varying degrees.Conclusion:Recuperating-heart granule can be used to prevent and cure the model of similar unstable angina pectoris.The result of this study indicate that it can decrease the content of PAPP-A,restrain endangium hyperplasia and improve pathomorphism of arteria carotis communis.To reduce aggregation of inflammatory cell interior the plaque and migration of smooth muscle cell,participate in the reaction of inflammation and stabilize the unstable atherosclerotic plaque.So it can prevent and treat unstable angina pectoris. |
