| ObjectiveThe important patho change in earlier -period diabetic retinopathy is the leucocyte adheres to the retina blood capillary and induces the breakdown of blood-retinal barrie and the capillary nonperfusion.This course is mediated by endotheliocyte secreting ICAM-1.There are findings have confirmed that the ICAM-1 conspicuous increasing and confirmed that the increasing of the immunocompetence has dependability with the competent of the blood sugar level.There are findings have displayed that if give earlier period masculine wistar applying Valsartan 12 weeks continuously can make that their pressure value step down conspicuously than control group.And their ICAM-1 of Aortic tunica intima conspicuous step down too,showing that AT1R not only can cut down blood pressure but also can reverse the damage of the blood vessel endothelium.Valsartan is a new type of non- peptid Angll acceptor-1 blocker(AT1R),this research in order to improve diabetic retinopathy (DR)by observing Valsartan can restrain the production of ICAM-1 so that improve DR.,it has significance in the prevention and curing of DR.MethodsA total of 115 healthy male Wistar rats 180-220g were used,40 were randomly chosen to be diabetic group,40 were randomly chosen to be diabetic+Valsartan group, and the rest 35 were randomly chosen to be normal group.Diabet group and diabetic+ Valsartan group was induced by streptozotocin(STZ) wich was injected into the abdomen,and normal group were injected tales doses citrate buffer solution.All the rats were normal bred and according to body weight and groups giving intragastric administration differently everyday.2,4and 6 weeks after rat modal of diabetes,executing 10 rats respectively.The imrnunohistochemistry method was used to measure the expression of ICAM-1 in the retina.ADP enzymohistochemistry stain method of retinal stretched preparation was used to observe the change of retinal blood vessels.Date was analyzed using the Statistical Package of the SPSS 12.0.Descriptive data were given as(?)±s.Two group continous variables were tested by analysis of T-test.Many group continuous variables were tested by analysis of one-way ANOVA and SNK test.The p-values are two-tailed and p-value of less than 0.05 being considered to be significant.Results1,In the experiment period the weights of the diabetic group and the diabetic +Valsartan group were lower than the normal group.2,Imrnunohistochemistry method showed that the positive staining particles of ICAM-1 in the retina were found in the normal group.In the diabetic group and the diabetic+Valsartan group the positive staining particles of ICAM-1 increased markedly.The differences of the expression were significant.The differences of the expression were significant in 2 week 4 week and 6 week(P<0.05).And between the diabetic group and the diabetic+Valsartan group the positive staining particles of ICAM-1 increased in diabetic group increased markedly than diabetic+Valsartan group in 2 week 4 week and 6 week(P<0.05).3,ADP enzymohistochemistry stain method showed that retinal vessels perfuse degrade and vascular occlusion were found in the diabetic group in 6 weeks than the the diabetic+Valsartan group and the normal group.The differences exist between the diabetic+Valsartan group and the normal group,but were not so significant.Conclusion1,The expression of ICAM-1 was much increased in the diabetic rats than the control rats,but the expression of ICAM-1 was much decreased in the diabetic+ Valsartan group rats than the diabetic group.2,In the retina of diabetes mellitus rats,the expression of ICAM-1 increase markedly.3,Intervening the expression of ICAM-1 is probably a new way of preventing and treating diabetic retinopathy. |
