The Relevance Research On Sex Hormones And Immune Factors (IL-4, IFN-γ) In Deficiency Kidney-Yang's Mice Induced Asthma

Purpose:The purpose of this study was to explore the nature of the micro-deficiency of kidney-YANG and the correlation of the level of sex hormonrs and immune factors in deficiency of kidney-YANG asthenia.TCM believe that the cause of the recurrent asthma is "deficiency of kidney-YANG".In recent years,many scholars study that deficiency of kidney-YANG is closely related to gonadal function and have changes in sex hormone levels. Clinical Medicine believe that Asthma's basic pathological changes is the airway hyperresponsiveness inflammation,but immune disorders is the main mechanism.This subject is combined with TCM of deficiency of kidney-YANG and Clinical Medicine of asthma,to study relationship of sex hormones(T) and immune factors(IL-4, IFN-γ).Significance:To further the substance of deficiency of kidney-YANG and to look for more clinical treatment of related diseases.Such as asthma,the asthma prevalence and mortality rates are still increasing year by year.Glucocorticoid treatment of asthma is the most effective means of side-effects,but many scholars dedicated to the search for non-hormone treatment.If the study confirmed the imbalance of sex hormone and immune dysfunction,stengthen kidney -YANG may will be a good means.Methods:The experimental male mice were randomly divided into five groups,the normal control group,the group of normal control and wheezing,the group of deficiency of kidney-YANG and wheezing, the group of deficiency of kidney-YANG and wheezing and goldkidney-QI pill,the group of deficiency of kidney-YANG and wheezing and BA Capsule.The model mice of deficiency of kidney-YANG was made by removing two sides testis and parastata,using aluminium hydroxide suspl with egg albumin(OVA) of peritoneal injection to allergize it,using the method of atomizating normal saline solution(NSS) with OVA and sucking in to arouse its asthma.During lab process,observing and taking notes of every mice group's change about food and drink,urination and defecation,spirit condition,color pattern,range-of-motion,body weight;carrying out statistical analysis to thirty minutes' symptom index of the last luring wheeze.Using radio-immunity(RI) to detect the blood content of serum testosterone(T).Using Enzyme-linked immunosorbent assay to detect the blood content of serum interleukin 4(IL-4),γ-interferon(IFN-γ).Simultaneously undertaking pathomor-phology observation of mice lung tissue.Results:1.In each group of deficiency of kidney-YANG,the mice had these symptoms like chilly,philo-warm,diuresis,thin and noncoagulative sedes,dull reaction,scrunch,olig-extrado,less to move and so on.In the group of goldkidney-QI pill,the symptoms above-mentioned became lighter,and discovered that the group of deficiency of kidney-YANG was easier to be lured to wheeze,and in it the symptoms abovementioned became significant.2.Through detecting the sexual hormone(SH) content,discovered that the T value in the group of deficiency of kidney-YANG and wheezing was significantly lower compared with normal controls(P＜0.01);the T value in the group of deficiency of kidney-YANG and wheezing and BA Capsule rised(P＜0.01);Compared with the group of deficiency of kidney-YANG and wheezing,the T value in the treatment group were higher(P＜0.05).3.Detection of immune factors found that the levels of IL-4 in the group of deficiency of kidney-YANG and wheezing and the group of deficiency of kidney-YANG and wheezing and goldkidney-QI rised notably compared with normal controls(P＜0.01).And the levels of IL-4 in the treatment group dropped(P＜0.01).the levels of IFN-γin the other group dropped compared with normal controls(P＜0.01). In the treatment group rised compared with the group of deficiency of kidney-YANG and wheezing(P＜0.05).4.Through undertaking pathomorphology observation of mice lung tissue the histopathology change became more obvious in the group of defic-iency of kidney-YANG lured to wheeze than in that normal control group lured to wheeze,after giving the traditional Chinese medicine of warm-ming kidney and pacifying wheeze,that histopathology change was just a little.Conclusion:1.Proved that the the formation of animal model of Yang Deficiency related to the level of changes sex hormones on the basis of senior research experiments. 2.To Prove that deficiency of kidney-YANG is one of important pathologic basis in bronchial asthma.3.Further confirmed that sex hormone changes in animals with the immune factors associated with inter-changes.Million and sex should be hormones,immune factors are its lower-level factor. Therefore,in accordance with TCM principle,use the linkage relationship between the two,in correcting abnormal sex hormone in patients with asthma and make it back to the premise of the normal level,you can adjust their immune imbalance factor.4.Provide a new idea in the treatment of bronchial asthma.
