ObjectiveTo observe the effects of Jiaweididang decoction on t-PA,PAI-1 content in blood and the expression of a-SMA,PAI-1 in kidney tissue of unilaternal ureteral obstruction(UUO) rats,and explore the mechanism of the effect.48 Wistar rats were randomly divided into four groups:model group,Jiaweididang group,Benazepril group,sham operation group.12 rats were in every group. Inject 30mg/kg 2%Pentoarbital Sodium in abdominal cavities of the rats of model group,Jiaweididan group,Benazepril group and sham operation group,and fixed them after they were anesthetized.Prepare skin;opened abdominal cavities;ligatured left ureters and sewed up the wounds.Every day gave every rat an intramuscular injection of 80 thousand units penicillin to prevent infection.Every day poured 1.8ml/kg physiology salt solution into the rats' stomachs of model group in 14 days.Jiaweididan group:every day poured stomachs with Jiaweididan decoction in 14 days,and dose was 1.8ml/(kg/d).Benazepril group:everyday poured stomachs once in 14 days,and dose was 1.8ml/(kg/d).Sham operation group:samely anesthetized and openedabdominal cavities,but didn't ligature ureters;one layer by one layer sewed up the wounds,gave an intramuscular injection of penicillin,every day poured stomachs with 1.8ml/kg physiology salt solution in 14 days.Excise eyes of every group rats on the fourteenth day,and gathered blood into test tubes.Delivered the blood to the Chechout Room of Hubei Province Traditional Medicine Hospital to measure the creatinine,Scr,BUN,t-pA,PAI-1content.Made the roots died;got the kidneys of ligation side and preserved them into 10%neutral ormalin to make kidney tissue into slices and measure the a-SMA content in them.After making kidney tissue into slices and being dyed by HE and Masson,observed and compare degree of renal interstitial fibrosis of various groups under high times lens(×400).Result:1.Determination of renal function:the sham group compares to other three groups Scr were increased after the Jiaweididang decoction and Benazepril treatment,the drop was not significant(P>0.05).Detection and the remaining three groups,BUN were significantly increased(P<0.05),but compared with the model group,the Chinese medicine group and Benazepil group were significantly decreased(P<0.05),the two treatment comparison between groups,there was no significant difference(P>0.05),After that treatment,Scr not obvioused decline,BUN has significantly decreased2.Renal pathology:the naked eyes observation that:sham operated rats kidney dark red,size,shape normal.model group, the jiaweididang decoction and Benazepril ligation kidneys of rats color desacination obviously enlarged,cystic sexy,memory tawny turbid liquid,pelvis obvious expansion,the real thinning. Optical microscopy:HE staining,sham-operated group occasionally tubular epithelial cells degeneration,kitchen-infict-rating mononuclear cells,model group that diffuse tubular epithelial degeneration,some of the lumon with protein or cell type,with small foci the expansion,contrction,occasionally necrosis,renal interstitial how stove monocyte infiltration and fibrosis,glomer rular no significant pathlogical changes,the jiaweididang decoction and Benazepril these changes reduce both.3.Area of Masson staining of renal interstitial fibrosisModel group that interstitial collagen fibers with Large blue proliferation,glomerular lesions not obvious,Chinese medicine group and Benazepril group renal interstitial collagen fibers positive staining area compared to the model group were signicantly decreased(P<0.05), between the two treatment groups,the difference was not statistically significant(P>0.05).4.Measure of t-PA and PAI-1 content in bloodContent of t-PA in Jiaweididang group was obviously higher than model group(p<0.05),and was lower than sham operated group(p<0.05).Jiaweididang group was quivalent to Benazepril group.Content of PAI-1 in Jiaweididang group was obviously lower than model group(p<0.01),and was taller than sham operation group.5.the expression of a-SMA in kidney tissue of every group sham-operated group compares to other three groups a-SMA were significantly increased(p<0.05),after Jiaweididang decoction and Benazepril treatment,the drop was signi ficant(P<0.05). sham-operated group of vascular smooth muscle cell wall was positive expression,and glomerulal no significant,renal tubular epithelial cells coloring,renal medualla were scattered in the sparse weak positive,model section of the site to renal inters -titial was mainly on the expansion or contraction of the tubular around,or renal interstitial fibrosis in Jiaweididang decotion and Benazepril group that is scattered renal interstitial,a small expression.6.the expression of PAI-1 inkidney tissue of every groupPAI-1 in renal tissue of sham-operated group,only a small number in the blood vessels and expression of renal tubular epithelial cells,no obvious glomerular expression.In the model group,PAI-1 expression in an area of significant increase, the expression of Jiaweididang decoction group than the model group was significantly weakened,with a considerable Benazepril group.Conclusion:The research indicated that Jiaweididang decction has the effection of preventing renal interstitial fibrosis.The mechanism probably be to enhance fibrinolytic activity of the rat,to promote the degradation of ECM,to mainten renal interstitial normal cycle.Prevention of renal ischemia resulting from interstitial nephritis.Jiaweididang decction can inhibit the expression of a-SMA and PAI-1 protein in renal tissue,Inhibit renal tubular epithelial cells and promote the degradation of ECM,so as to reduce the degree of renal fibrosis.
