| Objective: To observe the effect of Schwann cells on the expression of ciliary neurotrophic factor,and explore the protection of Schwann cells on the retinal ganglion cells.Methods: By Dual-enzyme digestion method isolated and cultured, purified rat Schwann cells;the intravitreous was injected with Schwann cells;three days later, to establish the acute high intraocular pressure model by the anterior chamber perfusion;After successfully established,using RT-PCR to detect the expressions of ciliary neurotrophic factor mRNA in the rat retina at different stages of the 1st day,3rd day,7th day,14th day and 21th day;by the Two-step immunohistochemical metheod to detect the different expression level of protein.Results: Successfully isolated and cultured rat Schwann cells;established an acute high intraocular pressure model.RT-PCR results showed that the ciliary neurotrophic factor mRNA expression was gratually enhanced after acute high intraocular pressure injury,while weakly in normal rat retina tissues,and reached the peak expression level in the 3-7th days of the injury,then began to decrease, ciliary neurotrophic factor mRNA expression changes in DMEM intervention group were similar to the acute hypertension goup, in the Schwann cells intervention group the ciliary neurotrophic factor mRNA expression was higher than both in the acute hypertension goup and the DMEM intervention group at each stage,there were significant differences(p<0.05).The Immunohistochemical staining matched with PT-PCR expression in retina slices,Positive staining product positioning showed,gangle cell layer had little ciliary neurotrophic factor positive product expression in normal rat retina,and inceased after acute ocular hypertension injury. Ciliary neurotrophic factor positive products could be found in gangle cell layer,nerve fiber layer and inner nuclear layer.Ciliary neurotrophic factor positive products were stained obviously in gangle cell layer,nerve fiber layer,inner nuclear layer,outer nuclear layer of the Schwann cells intervention group among the 3rd day ,7th day ,14th day.Conclution: Retinal ciliary neurotrophic factor expression level rised after Schwann cells were injected into the intravitreous of the acute high intraocular pressure rat,suggesting that Schwann cells might be important in the recover of the retinal and optic nervel injury. |
