Adsorption condition:pH 6,temperature 25℃,the flowing rate was 3ml/min;desorption condition:eluent is 2.5%NaOH aqueous solution,the flowing rate was 3ml/min.With the condition of optimum separation and purification,the adsorption rate was 83.35%,desorption rate was 93.65%,the yield of shikimic acid was 85.06%,the purity was increased from 6.15% to 21.76%with 3.6 times as high as the crude extracts.Research on the methodology of purification shikimic acid by column chromatography of silica qel.The concrete methodology condition was as follows:the filler of column chromatography was 100-200 silica qel;loading amount was 0.4g/10g silica qel;eluting solvent is acetic ether-aether petrolei-formic acid 7:2:1;the flowing rate was 3ml/min;getting the corresponding eluting solvent together.8g sample which was separated from extraction shikimic acid by column chromatography had 72.14%purity 1.5g and 78.21%yield. |
