| OBJECTIVE:To explore the effects of Interleukin-10(IL-10) on expression of signal transducer and activator of transcription3(STAT3) in pancreatic tissue and hepatic tissue of acute necrotizing pancreatitis(ANP) rats, and to investigate the expression of STAT3.METHODS:Ninty-two Sprague-Dawley male rats were divided randomly into three groups:normal control group(group C,Nï¼24),ANP group(group A, Nï¼36),IL-10 group(group I,Nï¼32).The model of ANP was induced by intraperitoneal injection(ip) of L-Arginine.Group I was treated with 10000 unites of recombination human IL-10(rhIL-10) by ip at the 2th,5th and 8th hour after the last L-arginine injection.Rats were killed at the 4th,12th,24th and 36th hour after the last L-Arginine injection.The level of serum amylase and the histopathological scores for pancreas and hepars were investigated,and the expression of STAT3 was detected by immunohistochemical(IHC) staining.RESULTS:1.blood serum:The levels of serum amylase of group A were significantly higher than group C(P<0.01) and group I(P<0.05) at different time points.2.pancreatic tissue:The histopathological scores in group A were significantly higher than group C at different time points(P<0.01),and significantly higher than group I at the 12th hour,(P<0.05),and maintained at high scores at the 24th and 36th hour(P<0.01);The histopathological scores in group I were significantly higher than group C at different time points(P<0.05). The expression of STAT3 of group A was significantly higher than group C at different time points(P<0.01),and significantly higher than group I at the 12th and 24th hour,(P<0.01),and maintained at high grey values at the 36th hour(P<0.05);The grey values in group I were significantly higher than group C at different time points(P<0.01).3.hepatic tissue:The histopathological scores in group A were significantly higher than group C at the 4th,12th and 24th hour(P<0.01),and maintained at high scores at the 36th hour(P<0.05),and significantly higher than group I at the 24th hour(P<0.05);The scores in group I were significantly higher than group C at the 4th,12th and 24th hour(P<0.01),and maintained at high scores at the 36th hour(P<0.05).The expression of STAT3 group A was significantly higher than group C at different time points(P<0.01),and significantly higher than group I at the 24th and 36th hour,(P<0.01).The expression of group I was significantly higher than group C at different time points(P<0.01).CONCLUSIONS:1.To a certain degree,the recombination human IL-10 that is intraperitoneal injected after the last L-arginine injection has an inhibiting and protecting effect on pancreatic tissue and hepatic tissue injury of ANP rats.2.STAT3 may participate in the early inflammatory response of acute necrotizing pancreatitis rats in pancreatic tissue and hepatic tissue,and may have a partial anti-inflammatory response effect through the endogenous IL-10/STAT-3.mediated anti-inflammatory response in pancreatic tissue. |
