Objective To study the effects of mangiferin on cell cycle status and cell cycle controlling genes expression in HL-60 cells , further to study the melocular mechanism for treating leukemia.Methods The change of cell cycle status in HL-60 cells treated with different concentration of mangiferin for 24 and 48 hours was performed by the flow cytometric ; The expression of CDC2,CyclinB1,CyclinA,Chk1,CDC25C and Wee1 mRNA was detected by semiquantitative RT-PCR.Results Mangiferin could induce G2/M phase arrest on HL-60 cells.HL-60 cells in G2/ M phase increased in a dose-dependent way after treatment with mangiferin for 24 hours, and the proportion of which with 160μmol·L-1 mangiferin was significantly higher than that occurring in untreated cells (28.9&5.2%,P<0.01),indicating G2/ M phase blockage ; Also the proportion of cells in the G2/M phase after treatment with 160μmol·L-1 mangiferin for 48 hours was lower than that occurring treated cells for 24 hours (15.3&28.9,P<0.01);The expression of CDC2,CyclinB1,CyclinA,Chk1,CDC25C and Wee1 mRNA enhanced after treament with mangiferin for 24 hours.Exposure of HL-60 cells from 20 to 80μmol·L-1 for 24 h had higher expression of CDC2,CyclinA,CDC25CmRNA than that occurring treated with mangiferin(≥160μmol/L)(P<0.01),and exposure of HL-60 cells from 20 to 80μmol·L-1 for 24 h had lower expression of CyclinB1,Wee1mRNA than that occurring treated with mangiferin(≥80μmol/L)(P<0.01). It's not statistical significance that expression of all genes treated with mangiferin for 48 hours than control(P>0.05).Conclusion :In the earlier period mangiferin can blocked HL-60 cell cycle progression in G2/ M phase ,and it can significantly increase the expression of CDC2,CyclinB1,CyclinA,Chk1,CDC25C and Wee1 at the mRNA level in HL-60 cells , G2/ M phase blockage may be one of its molecular mechanism for treating leukemia. |
