| Objective:To investigate the therapeutic effects of angiotensinⅡtype 1 receptor antagonist(valsartan) on rats renal interstitial fibrosis caused by unilateral ureteral obstruction nephropathy(UUO).Methods:Wistar rats were used to establish the UUO model,and then they were divided into the following three groups,,control group,model group,valsartan group(30 mg·kg-1·d-1).Rats were sacrificed 3,10,20 days after obstruction.Scr and BUN were measured by Automatic Chemistry Analyzer.The expression of transforming growth factor-β1(TGF-β1) mRNA was semi-quantitatively determined with reverse transcriptionpolymerase chain reaction(RT-PCR).The expression of OPN was analyzed by immuno-histochemistry.Results:Compared with CTR,Scr and BUN were significantly increased since the 10th day after operation(P＜0.05 or 0.01),immuno-histochemistry revealed that the expression of OPN and the expression of TGF-β1 mRNA were significantly up-regulated in the model group(P＜0.01). Compared with UUO,after intervention with valsartan,Scr,BUN,OPN,and the expression of TGF-β1 mRNA were significantly down-regulated.the up-regulating above mentioned parameters were all significantly inhibited(P＜0.05 or 0.01).Conclusions:The expression of OPN and TGF-β1 mRNA increased in the model group with the time going,suggested that OPN and the expression of TGF-β1 mRNA have relationship with the progression of renal interstitial fibrosis.After intervention with valsartan,BUN,Scr,the expression of OPN and TGF-β1 mRNA decreased visibly compared... |
