Objectives To investigate the effects of synthetic adrenocorticotropic hormone (ACTH1-24,ACTH4-10) on serum lipid levels,renal function and the adrenal gland in normal NZW rabbits and hyperlipidemic rabbits.Methods The study included two parts⑴Thirty rabbits were randomly assigned to receive an intramuscular injectionn of ACTH1-24 at a dose of 100μg/kg(ACTH1-24 group n=10),ACTH4-10 at a dose of 300μg/kg(ACTH4-10 group n=10) and the same volume of saline solution(control group n=10). Serum levels of lipid and renal function were detected while adrenl status was assessed.⑵Thirty hyperlipidemic rabbits were randomly assigned to receive subcutaneous injections of ACTH1-24(100μg/kg), ACTH4-10(600μg/kg) and the same volume of saline solution. Serum levels of lipoproteins, renal function were detected and adrenl status was assessed.Results⑴Study in rabbits feed normal diet: Compared with control group, serum levels of TG, TC and LDL-C were markly reduced and renal function was imposed while adrenal growth was evident in the ACTH1-24 group. There was no changes in ACTH4-10 group.⑵Study in hyperlipidemic rabbits: Compared with rabbits feed nomal diet, the levels of TC, TG, LDL-C and HDL-C markedly increased in the hyperlipidemic rabbits, while the renal function was impaired. Serum levels of TC,LDL-C and HDL-C were significantly decreased , but the level of TG was markly increased and the impaired renal function was imposed in response to ACTH1-24 treatment. In ACTH4-10 group, we found that the levels of BUN was markly decreased, while the HDL-C was rather increased.Conclusion The NZW ribbit model is suitable for the research on the mechanism of action of ACTH on lowering lipids. ACTH1-24 can improve the impaired renal function due to hyperlipemia. ACTH4-10 may also play a role in serum lipid metabolism, but there is some difference compared with ACTH1-24. ACTH4-10 can not induce growth of the adrenal glands and decrease the body weight of rabbits.
