Objective:The use of animal experiments, through the X-ray irradiation SD rat eyes set up animal model of radiation cataract were observed by X-ray radiation dose and the severity of cataract relations; Detected by different doses of X-ray irradiation of rat lens solubleαB-crystallin the expression level ofαB-crystallin to explore with the radioactive relationship between cataract formation.Methods: Select 8 pre-irradiation of rats as normal control group, experimental exposure to rats 24, the average is divided into 3 groups each group 8, one-time irradiation, respectively (both eyes simultaneously irradiation) 5,15,25 Gy. Experimental control group of 24 were divided into 3 groups each of 8, 5,15,25 Gy, respectively, as a control group of sham irradiation (irradiation of light in the non-radiation, the other conditions were the same as corresponding to the irradiated group). Rats were randomly check each eye for the study as the experimental eye. All rats were irradiated 1,3,5,7,15,30,45,60,90 d, intraperitoneal injection of 3.6% chloral hydrate anesthesia, the use of prescribed eye drops to fully mydriasis, slit-lamp microscope and record the lens opacities case; in 3 months after irradiation the rats were sacrificed, check the lens for homogenate, select the supernatant (soluble lens protein) used immunoblotting (Western blot) detection ofαB-crystallin expression.Results: The observation of 3 months, normal control group, the experimental control group and 5 Gy rats no abnormal lens. 15 Gy group 45 d after irradiation and after posterior capsular cortex began to appear small, scattered spots in the white turbidity, and then blended into more labor-intensive, non-uniform punctate, patchy opacities, the further development of a more even after the emergence of dense cortical opacities, the former cortex , anterior capsule opacification and mild core. One of 3 months up to 62.5% in stage Ⅱ, 37.5% for stageⅢTatsu. 25 Gy group similar changes, but the turbidity of time crystal happened sooner, faster, more obvious turbidity. 30 d after irradiation that is, posterior capsular opacification spots began to appear, since the rapid development of lesions, to 45 d when there is 37.5% of all crystal turbidity. To 3 months time in addition to Example 1 under the clear posterior capsule opacification, the remaining seven cases are fully turbid lens; experimental control groupαB-crystal between the soluble protein content was no significant difference (P> 0.05), the experimental irradiated group of solubleαB-crystal protein content than the control group decreased significantly (P <0.05), 3 experimental irradiatedαB-crystal soluble protein content with increasing doses gradually reduced, the difference has significant statistical significance, (P <0.05).Conclusions: High-dose X-ray in the short term can cause lens opacities; radioactive cataract formation of the speed and extent of radiation dose and a positive correlation at radiation cataract in the lens inside the soluble expression ofαB-crystallin reduced cataract formation play an important role.
