Partâ… Study on mRNA and protein expression changes of NSE and GFAP in the reduced glutathione-induced differentiation process from rat bone marrow stromal cells to neuron-like cellsObjective:To investigate the induced differentiation of rat bone marrow stromal cells with reduced glutathione(GSH).Method:Rat BMSCs were cultured and purified in vitro.Afterwards,GSH was administrated to induce BMSCs to differentiate into neuron-like cells.After 6h,12h and 24h,real-time PCR and Western blot were applied to detect the mRNA and protein expression levels of NSE and GFAP.Results:After being treated with GSH for 6h,12h and 24h,BMSCs were observed to become neuron-like cells and glia-like cells.Real-time PCR and Western blot experiments showed that the mRNA and protein expression levels of NSE and GFAP were significantly higher in the GSH group than those in the control group.This difference was most remarkable after BMSCs were induced for 12 hours.Conclusion:GSH may induce BMSCs to differentiate into neuron-like cells and glia-like cells,in accordance with increased mRNA and protein expression levels of the NSE and GFAP,respectively. Partâ…¡Preliminary study on the gene expression profile associated with in vitro EGF and bFGF-induced differentiation of rat bone marrow stromal cells into neuron-like cellsObjective:To detect the expression profile of interesting genes,especially of neuron development genes,involved in the EGF and bFGF-induced differentiation of bone marrow stromal cells(BMSCs) into neuron-like cells in vitro.Method:In vitro cultured and purified rat BMSCs were induced to differentiate into neuron-like cells with bFGF and EGF.Before and 7 days after the inducement,rat Affymetrix expression microarrays were applied to detect the changes of key gene expression which was further analyzed with advanced bioinformatics techniques.Results:216 differently expressed genes were detected,among which 125 were up-regulated and 91 were down-regulated.In this group,13 genes(7 up-regulated and 6 down-regulated) in the literature reported to be closely related to neural development were specially identified.Conclusion:BMSCs may differentiate into neuron-like cells.13 important genes closely related to neural differentiation were specially expressed.These genes were no doubt good candidates for further study.
