Objective:This study aims to explore the effect of acupuncture on proteome in L5 spared dorsal root of SD rat deafferentated.Method:SD rats were divided into 4 groups randomly,known as sham operation group,deafferentated group,acupuncture group,electro-acupuncture group Respectively.At 14day post operation(dpo),L5 DRG from each group was harvested. After determined the protein density by Bradford,The samples from each group were separated by 2-DE and subjected to silver staining,maps of proteins were analyzed using PDQuest software.Lastly,the proteins were performed by MS/MS identification and bioinformatics analysis.Result:There medially are 500 protein spots in the gels.We respectively find 14 differential spots between sham-operation and 14dpo,14dpo and 14Da,14dA and 14d EA.14 differential protein spots are excised and identified by using mass spectrometry(MS) and MS/MS,named as C-reactive protein,Immunoglobulin lambda- light chain,Lectin galactose binding soluble 5,Fast myosin alkali light chain- If,Chainl alpha-parvalbumin,α-B-crystallin,Myosin light polypeptide 2,Fast myosin alkali light chain -3 f,β-galactoside-binding lectin,Enolase 2γ.Among them, 4 proteins in Acupuncture group were up regulated than in operated group.Whereas, 1 protein was up regulated in EA group than acupunctured group.Conclusion:This study shows some changes of proteome in spared dorsal root ganglia of rats subjected to acupuncture.It provides some evidences for understanding the positive effect of acupuncture on spinal cord injury in molecular level.
