Optical clearing technique provides a new break point for obtaining deeper structural information of tissue. However, most researches focus on the optical clearing effect of local area of tissue, less attention is paid to the changes of hemodynamic parameters, such as the velocity of blood and the diameter of vessels, which reflect the function of tissue by using optical clearing agents.In this paper, Laser speckle imaging technique is applied in real-time monitoring on the changes of velocity of blood and the diameter of vessels induced by optical clearing agents by using the chick chorioallantoic membrane (Embryo age 6~15) model. The optical clearing agents were glycerol of different doses in different densities, so as to glucose solutions.The results show that:(1) The glycerol or the glucose has some common effects on vessels. The effect of the velocity of the blood flow and the diameter of vessels are in direct proportion to the concentration and the dose.(2) It is notable that the glucose seems to have prolonged effect, and the effect of glycerol is rapid.(3) The effect of each agent on blood microcirculation in a long period of time has distinctive features to each other. The vessels are distorted after applying the glycerol whose blood flow is still existed while the blood of this kind of vessels kept stasis after the application of glucose.(4) Both the glycerol solutions and the glucose solutions have no effects on the blood flow and vessels at low concentrations and less doses.The study on the effect of optical clearing agents to the vessels is potentially available for acquiring the exact information of the vessels, as well as the investigation on the biocompatibility of different agents aiming at the choice of the safest optical clearing agents, which is significantly to the development of the optical clearing technique.
