Immunohistochemical Study Of The HuD Protein Of The Colon In Hirschsprung's Disease And Hypoganglionosis

Objective: HuD protein involved in neurite growth and mature neurons survival is the necessary factor in the embryonic and adult development nervous system.It is not clear that the role of HuD in developing enteric nervous system and pathogenesis of Hirschsprung's Disease and Hypoganglionosis.The study of the expression of HuD protein in the bowel segments of Hirschsprung's Disease and Hypoganglionosis,is could help us to understand the relationship between HuD protein and enteric nervous system(Hirschsprung's Disease ,Hypoganglionosis).Methods:To get 15 samples from Hirschsprung's disease and 15 samples from Hypoganglionosis,and 6 samples from patients with no enteric nervous system disease.Using immuocytochemistry to determin the expression of HuD protein in the bowel segments.To analysis the result by image dissector systemResults: The stain of anti- HuD protein was negative in the segments of the Hirschsprung's disease.The stain of anti- HuD protein in the segments of the Hypoganglionosis was low than the normal control groups. Conclusions: The HuD protein in the submucosa and intermuscular nervous plexus was negative or low expressed,it suggest that decreasing of HuD protein could cause nervous system embryodevelopment or matural neuron surival disorder.The last ,this disorder may lead to the the abnormal of the enteric nervous system,i.e: Hirschsprung's Disease and Hypoganglionosis.
