Part one Expression of B7-H1 on Peripheral Blood White Cells from the Patients with Systemic Lupus ErythematosusObjectives: To investigate the expression of B7-H1 on peripheral blood White cells(PBWCs) from the patients with systemic lupus erythematosus (SLE) and its role.Methods: Immunofluorescent staining and flow cytometric assay were used to analyze the level of B7-H1 expression on PBWCs from 30 SLE patients and 20 healthy volunteers. The data of Systemic Lupus Erythematosus Diseases Activity Index (SLEDAI) in SLE patients were collected.Results: The level of B7-H1 expression on monocytes,lymphocytes and granulocytes from active SLE group was markedly higher than that in the control group (P<0.01). There was positive correlation between B7-H1 molecule on monocytes,lymphocytes and SLEDAI (r1=0.418, P1<0.05; r2=0.519, P2<0.05). On the other hand, B7-H1 molecule expression on lymphocytes from the patients with positivity anti-dsDNA antibody was high, compared with that of patients with negative anti-dsDNA antibody (t=3.204, P<0.01). The level of B7-H1 on monocytes, lymphocytes and granulocytes was obviously decreased after the disease was controlled (t1=9.977, P1<0.01; t2=6.753, P2<0.01; t3=4.326, P3<0.01).Conclusions: The expression of B7-H1 on PBWCs from active SLE is increased, and there was positive correlation between B7-H1 molecule and SLEDAI, which might play a role in the pathogenesis of SLE.Part two Expression of B7-H1 and PD-1 on PBMCs from the patients with Systemic Lupus Erythematosus Objective To investigate the expression of PD-1 and B7-H1 on PBMCs from the patients with systemic lupus erythematosus (SLE) and its role.Methods Immunofluorescent staining and flow cytometric assay were used to analyze the level of B7-H1 expression on PBMCs from 25 SLE active patients and 20 SLE inactive patients and 20 healthy volunteers. The data of systemic lupus erythematosus diseases activity index (SLEDAI) in SLE patients were collected.Results①The level of PD-1 expression on monocytes, lymphocytes from active SLE group was markedly higher than that in the control group (P<0.01).②There was positive correlation between PD-1 molecule on monocytes, lymphocytes and SLEDAI (r1=0.915, P1<0.05; r2=0.841, P2<0.05).③The level of B7-H1 expression on CD3+T cells, CD19+B cells from active SLE group was markedly higher than that in the control group (P<0.01).④There was positive correlation between B7-H1 molecule on CD3+T cells, CD19+B cells and SLEDAI from active and inactive patients (P<0.05).⑤B7-H1 molecule expression on CD3+T cells and CD19+B cells from the patients with positivity anti-dsDNA antibody was high, compared with that of active patients with negative anti-dsDNA antibody (P<0.05).Conclusions The expression of PD-1 and B7-H1 on lymphocyte from active SLE is increased, and there was positive correlation between B7-H1 and PD-1 molecule and SLEDAI, which might play a role in the pathogenesis of SLE.
