| Rhizoma Curcumae, the rhizome of Curcuma phaeocaulis Val.,Curcuma Kwangsiensis S.G.Lee et C.F.Liang or Curcuma wenyujin Y.H.Chen et C.Ling, which is multi-origin variety and root in the family of Zingiberaceae. Their complex origin, wide distribution, unstandard cultivation and elaboration lead to its unstable quality, also with no effective quality evaluation method, made its quality control difficult. The work of this topic is part of the Tackling-key Project of Zhejiang Department of Science and Technology: GAP of C. wenyujin and new drug exploitation, mainly study the variety and quality of Rhizoma Curcumae from plant morphous, material character, micro-character, chemical composition, finger print.From the study on morphological and histological, we find the difference on plant morphous, material character, micro-character. The color of main rhizome and root tuber transection, with or without non-glandular hair and growth position, expulsion position of inflorescence are distinct in plant morphous. Size, surface character and section color are different in material character. The color of Oil cell, the number of non-glandular hair, leaf-trace bundle and vascular bundle are distinct in micro-character.On chemical composition aspect, essential oil, ligarine extractive, methanol extract are compared through TLC. Checked out absorbance, total ash, acid-insoluble ash according to Chinese pharmacopeia, increased the detection of moisture content, heavy metals, pesticide residue, also determined the content of essential oil, ethanol-soluble extractive. The assaying methods of total curcumines, curcumine, germacrone, curdione are established. The detection result indicated that the category and content of their chemical composition are notable difference. Total ash and acid-insoluble ash coincidence to pharmacopeia define.except part of guangxi ezhu, absorbance and essential oil coincidence to pharmacopeia define, furthermore essential oil content of wen ezhu is two times more than pharmacopeia define. It is suggested that add moisture detection, raise essential oil content standard, increase germacrone and curdione assaying of wen ezhu, curcumine, germacrone and curdione assaying of peng ezhu.On finger print aspect, we did some initial study of Rhizoma Curcumea through HPLC, and established the chemical composition finger print of wen ezhu and peng ezhu but guangxi ezhu because of its low correlation coefficient. The common finger print of the three species can't be established because of its low coefficient correlation.Through the research of chemical composition and finger print, also with report of pharmacology study and clinic use, we suggest that the three species should be solo use in pharmacopeia.The finding supplied scientific theory foundation for variety certification, quality evaluation, quality criteria establishment and construction of wen ezhu GAP depot. |
